Project description:The testis is one of the most transcriptionallyactive organs, however how germ cell differentiation is controlled at the molecular level remains largely unknown. To study global transcriptional dynamics during human spermatogenesis we sequenced total RNA of human testicular biopsies with 5 specific histological phenotypes: Sertoli cell-only (SCO, n=3), spermatogenic arrests at the spermatogonial (SPG, n=4), spermatocyte (SPC, n=3), and spermatid (SPD, n=3) level, as well as normal spermatogenesis (Normal, n=3). To our knowledge, this work provides the most complete capture of the transcriptome and its dynamics during human spermatogenesis to date.

Project description:In the present study we carried out testicular gene expression profiling of patients with AZFc microdeletion, idiopathic infertility and normal spermatogenesis, in order to identify up- and downregulated genes in the different conditions, and to verify the presence of specific molecular pathways leading to the spermatogenic damage in patients with AZFc deletions. All samples of patients with AZFc deletion clustered together, showing downregulation of several genes related to spermatogenesis. In this study, we analyzed testis expression profiles of 16 patients with different phenotypes, including men with AZFc deletions and men with idiopathic infertility. All pathological human testis RNAs were hybridized against normal testis reference RNA samples (obstructive azoospermia), while the three samples with normal spermatogenesis were hybridized against a control represented by an home-made RNA universal reference consisting of a commercial human purified tissue RNA pool (brain, liver, muscle and lung) (Clontech, Terra Bella, CA, USA) . These latter hybridizations were carried out in order to generate a profile of genes specifically expressed in normal testis.

Project description:Normal, premalignant and various histological subtypes of testicular germ cell tumor (TGCT) tissues were hybridized against Universal Human Reference RNA (Stratagene) onto Agilent 60mer oligo microarrays (GEO accession no GPL885). In vitro time series of two TGCT cell lines, NTERA2 and 2102Ep, treated with retinoic acid for 0, 3, and 7 days were also included. The data set (30 hybridizations) is particularly useful for comparisons between various histological subtypes of TGCT versus each other or versus normal testis. Project description:The successful proliferation and differentiation of testicular cells are vital for the survival, continuity and preservation of species. However, a comprehensive molecular orchestration underlying the spermatogenic process in teleost testis development throughout the annual cycle remains elusive. In this study, the testicular cells derived from adult black rockfish at distinct stages—Regressed, Regenerating, and Differentiating were dissected via single cell transcriptome sequencing. A continuous developmental trajectory of spermatogenic cells, from spermatogonia to spermatids, was delineated, elucidating the molecular events involved in the spermatogonia development. Subsequently, the dynamic regulation of gene expressions associated with spermatogonia proliferation and differentiation was observed across spermatogonia subgroups and various developmental stages. A bioenergetic transition from glycolysis to mitochondrial respiration of spermatogonia during annual developmental cycle was demonstrated, and a deeper level of heterogeneity and molecular characteristics was revealed by re-clustering analysis. The developmental trajectory of Sertoli cells in annual reproductive cycle was examined, and the divergence of Leydig cells and macrophage were explored. Additionally, the interaction network between testicular micro-environment somatic cells and spermatogenic cell were established. Overall, our study provides detailed information on both germ and somatic cells within teleost testis during annual reproductive cycle, along with insights into the molecular regulation of spermatogonia dynamics, which lay the foundation for spermatogenesis regulation and germplasm preservation of endangered species.

Project description:In this work, we performed single cell RNA sequencing in spermatogenic cells without CTCF. We uncovered defects in transcriptional programs that explain the severity of the damage in the produced sperm. At early stages of spermatogenesis, transcriptional alterations are mild. As germ cells go throughout the specialization stage or spermiogenesis, transcriptional profiles become more altered. We found spermatid defects that support the alterations in the transcriptional profiles, and thus we conclude that CTCF depletion alters several transcriptional profiles mostly during spermiogenesis. Our data highlights the importance of CTCF at the different stages of spermatogenesis.

Project description:We investigated the transcriptional changes in human testis in the presence of AZFc/AZFb+c microdeletions. As one encounters a dominating 'germ cell effect' in this pursuit, testicular biopsies were pre-selected by clustering based on similar quantitative germ cell composition. Statistical testing was employed to filter genes that are influenced by the 'deletion effect' using the selected subset of biopsies, thereby obtaining several hundered genes that may be involved in YCMD-induced spermatogenic impairment.

Project description:Luteinising hormone (LH) is a key regulator of male fertility through its effects on testosterone secretion by Leydig cells. Mice in which the LH receptor is knocked out (LuRKO) show reduced testicular size, reduced testosterone, elevated serum LH, and a spermatogenic arrest that can be rescued by administration of testosterone. This study examines the onset of spermatogenic arrest in LuRKO males using transcriptional profiling of developing mutant and control testes. We also examine the initial stages of testosterone rescue of the phenotype, in order to identify key upstream regulators of testosterone-dependent spermatogenesis.

Project description:We investigated testicular biopsies from 28 men which presented with full spermatogenesis (Johnsen Score 10, 12 samples), arrest at the spermatid stage (Johnsen Score 8, 6 samples), arrest at spermatocyte stage (Johnsen Score 5, 5 samples) and Sertoli-cell-only syndrom (Johnsen Score 2, 5 samples).
