Murine mesenchymal stem cells undergo molecular changes after a single passage in culture

Project description:The properties of MSCs can be affected by long term culture, therefore casting doubt over the ability of late passage MSCs to accurately recapitulate their biology. Additionally, assumption within the scientific community is often made that early passage MSCs are still representative of the primary MSC population, however, little research has been done to support this. We compared the transcriptomic profiles of murine MSCs freshly isolated from the long bones to MSCs that had been expanded in culture for 10 days. Interestingly, we identified that a single passage in culture induced extensive alterations to MSC molecular signatures associated with cell cycling, differentiation and immune response.

Chemically defined, serum-free and xeno-free generation of mature type 2 alveolar epithelial cells from hiPSCs

Project description:HiPSCs were differentiated into alveolar type 2 cells (AT2s) following a stepwise differentiation protocol, mimicking lung development, using a chemically defined, serum-free, and xeno-free differentiation protocol. Samples contain biological triplicates of each developmental stage (hiPSCs, definitive endoderm, lung progenitor stage, passage 0 AT2s, early and late passage AT2s) using the same cell line. Additionally, a biological triplicate for a cell line from an idiopathic pulmonary fibrosis patient with a homozygous surfactant protein C mutation was done for late-passage AT2s comparison.

Transcriptome analysis of isogenic Neural stem cell for LRRK2 G2019S mutation.

Project description:We report the transcriptome data produced from isogenic neural stem cells with normal LRRK2 gene, or LRRK2 G2019S mutation. After gene correction from patient-derived iPSCs with LRRK2 G2019S, the isogenic lines were diffrentiated toward neural stem cells. To reflect on aging effect, theses cells were harvested to the early (passage number 1) or late stage (passage number 13), without replicate. Also, they were prepared at the early stage (passage number 3), triplicate.

D4 and C2 assessed for chimerism and germline transmission

Project description:Two independent ES cell lines (D4 and C2) were assessed for chimerism and germline transmission. Although these lines were able to produce chimerism at early passage, no germ line transmission was seen. After passaging in a proprietary medium and subcloning by detaching loosely connected cells and culturing in the recommended medium, both lines experienced an increase in potential as evidenced by an enhanced ability to go germ line. Analysis of early passage, and late passage detached and attached cells will allow the determination of genes that may be implicated in the increase of potential seen in these cells. EPC senescence

Project description:Transcription profiling of human umbilical cord blood derived Outgrowth Endothelial Cells (OECs) at early and late passages. Outgrowth endothelial cells were isolated from the mononuclear fraction of umbilical cord blood and cultured on collagen coated flasks. Once OEC colonies emerged they were expanded in culture to study cell growth kinetics. Total RNA was extracted from OECs at an early passage and OECs at a late passage which were becoming senescent . The purpose of this experiment was to were compared early and late passage OECs to determine how senescence affects the gene expression profile of OECs.

A Global Transcriptome Analysis of BMSC Senescence

Project description:Bone marrow stromal cells (BMSCs) can be expanded by serial passage, but expansion is limited by cell senescence. The nature of changes associated with BMSC serial passages was assessed. Transcriptome analysis of 10 early and 15 late passage samples from 5 subjects revealed 2193 differentially expressed genes; those highly expressed in early passage cells were overrepresented in skeletal system development, embryonic morphogenesis, tube morphogenesis, etc, while those highly expressed in the late passage BMSCs were overrepresented in nucleosome assembly; chromatin assembly, DNA packaging, etc. 57 BMSC samples from 7 donors were further analyzed for the transition from an early to late passage; 155 genes were highly correlated with BMSC senescence and a set of 24 genes was predictive of BMSCs lifespan. The change from an early to a late passage molecular signature occurred between passage 3 and 5. In contrast, senescence associated beta-galactosidase staining began to increase after passage 6 or 7 and colony formation efficiency began to fall after passage 7. These data indicated that the onset of molecular changes associated with BMSC passage varied among individuals and preceded changes in commonly used indicators of BMSC senescence. The set of 24 BMSC lifespan predictive genes will be useful in assessing the quality of clinical BMSC products.
