Project description:<p>We generated a collection of patient-derived pancreatic normal and cancer organoids. We performed whole genome sequencing, targeted exome sequencing, and RNA sequencing on organoids as well as matched tumor and normal tissue if available. This dataset is a valuable resource for pancreas cancer researchers, and those looking to compare primary tissue to organoid culture. In our linked publication, we show that pancreatic cancer organoids recapitulate the mutational spectrum of pancreatic cancer. Furthermore, RNA sequencing of organoids demonstrates the presence of both transcriptional subtypes of pancreas cancer.</p>

Project description:The goal of this experiment is to characterize the different cancer associated CAF subtypes that can be induced by tumour-derived signlas in pancreatic stellate cells (PSC). Co-cultures of PSC and tumour organoids were seeded in matrigel and grown for 6 days in basal media. Mono-cultures of PSC were used as controls. The PSCs were recovered by flow cytometry and subjected to single cell RNA-seq profiling via the ddSeq-Surecell system.
Project description:This dataset presents the gene expression profiles of 36 frozen pancreas samples. RNA from 9 normal pancreas samples, 9 samples from patients with chronic pancreatitis, 9 samples from patients with pancreatic cancer and 9 samples from patients with metastatic pancreatic cancer was extracted and hybridized individually to Affymetrix HG_U95Av2 microarray chips. The data present a gene expression profile of the pancreas in normal and diseased states and has been used to identify genes which may play an important role in disease of the pancreas.
Project description:This single cell RNA-seq experiment was performed to quantify DLL3 expression in tumor cells in small cell lung cancer patients.Tumors were rapidly dissociated after the surgical procedure using the Miltenyi Biotec Human Tumor Dissociation kit (cat# 130-095-929). Libraries were constructed using the VDJ NextGEM v1.1 10x Genomics Chromium kit according to the manufacturer's instructions. Samples were sequenced on a NextSeq 550 sequencer (Illumina).
