Single cell sequencing: Capturing the origin and dynamics of chromosomal copy-number heterogeneity

ABSTRACT: Shallow whole-genome sequencing of single cells from individual clonal (patient-derived colorectal cancer) organoids was performed to investigate the degree of copy-number heterogeneity that emerges and propagates during clonal organoid outgrowth of a single cancerous cell. Selected single cells were sequenced with higher coverage. In addition, daughter cells were isolated to directly link mitotic chromatin errors to chromosomal copy number changes.
