FAM50A Disruption in TOV21G Cells RNAseq

ABSTRACT: In this project we aimed to assess the transcriptional consequences of disrupting FAM50A in TOV21G cells in vivo. To do this we generated a dox inducible FAM50A gRNA allele such that we could conditionally inactivate FAM50A by feeding mice a diet containing doxycycline. Tumour masses were harvested from mice when they reached the ethical limit and RNA was extracted for RNA-Seq analysis.

Project description:We utilized inducible transgenic mouse with spontaneous lung cancer formation driven by mutant EGFR (by Doxycycline induction) for experiments. Mice fed with regular diet (RD) or high-fat diet (HFD) were induced lung cancer followed by tumor burden evaluation. Lung tissues were harvested for whole-genome transcriptomic, gene enrichment analysis, and cytokine array for potential signatures identification.Gene set enrichment analysis showed the whole-genome transcriptome in HFD vs. RD treated lung cancer was enriched in immune responses and cytokine stimulus.

Project description:COG-N-519 primary neuroblastoma cells were innoculated by subcutaneous injection into NSG mice. When tumours reached a size of 100-200mm3 miRNAs conjugated to nanoparticles were injected intratumourally every 2 days for 1 week. 24 hr after the last injection, snap frozen tumour tissue was harvested for RNA extraction and RNA-seq analysis.

De novo construction of T cell compartment in humanized mice engrafted with iPSC-derived thymus organoids

Project description:Hematopoietic humanized (hu) mice are powerful tools for modeling the action of human immune system and are widely used for preclinical studies and drug discovery. However, generating a functional human T cell compartment in hu mice remains challenging, primarily due to the species-related differences between human and mouse thymus. While engrafting human fetal thymic tissues can support robust T cell development in hu mice, tissue scarcity and ethical concerns limit their wide use. Here, we describe the tissue engineering of human thymus organoids from inducible pluripotent stem cells (iPSC-thymus) that can support the de novo generation of a diverse population of functional human T cells. T cells of iPSC-thymus engrafted hu mice could mediate both cellular and humoral immune responses, including mounting robust proinflammatory responses upon TCR engagement, inhibiting the growth of allogeneic tumor grafts and facilitating efficient Ig class switching. Our findings suggest that hu mice engrafted with iPSC-thymus can serve as a novel animal model to study human T cell-mediated immunity and accelerate the translation of animal studies into the clinic.

Expression of obese mice subjected to diet induced weight loss for 60 days

Project description:We used microarrays to study the gene expression dynamics during caloric restricted weight loss in obese mice after feeded with high fat diet. C57BL/6J male mice were placed on a high fat diet at 10-11 weeks of age in individual cages and kept on the diet until they reached ~ 41 g. After all the baseline (41g) measurement were made, each mouse was fed 75% of their ad libitum food intake in two divided daily "meals". The high fat diet was maintained.
