Project description:Gallus gallus avian eggshell is a biomineral composed of 95% of calcium carbonate on calcitic form and of 3.5% of an extracellular organic matrix (proteins, polysaccharides and proteoglycans). The calcification process occurred in the lumen of the uterus into the acellular uterine fluid, and is divided in three stages (initial, growth and terminal), where the concentration and the nature of proteins varies depending on stages. We have used mass spectrometry methodologies to identify and quantify proteins secreted into the uterine fluid proteins during the three stages of biomineralization.

Project description:Tamoxifen treatment in breast cancer patients increases their risk of developing uterine cancer that is more likely of poor prognosis. However, the exact mechanism leading to this increased and the worse outcomeendometrial cancer risk following tamoxifen exposure are still unknown, and effective preventive strategies are lacking. To address this question, we performed whole-exome sequencing of 21 (discovery cohort) and digital droplet PCR for 40 (validation cohort ) tamoxifen-associated uterine cancers. We identified genomic features that have been previously reported for uterine cancer, including recurrent mutations in PTEN, KRAS, TP53 and ARIDA1 and copy number alterations in the RTK and PI3K pathway. However, we also found that tamoxifen-associated uterine cancers have a substantially low frequency of PIK3CA and PIK3R1 mutations, the most prevalent mutations in non-tamoxifen-associated uterine cancers. Moreover, overall survival is significantly worse in uterine tumors without PIK3CA mutations, which could explain the observed poor prognosis in tamoxifen-associated uterine cancers . We also performed in vivo studies of normal uterine tissue and show that tamoxifen activates the PI3Kinase pathway and increases proliferation that in turn is abrogated by the PI3Kinase inhibitor, alpelisib. Together, this study uncovers the unique molecular profile of tamoxifen-associated uterine cancer and , offers mechanistic insights in the observed increase in endometrial cancer risk following tamoxifen treatment and a potential preventive approach. Moreover, we here describe for the first time a new general principle that a drug, as a side effect, can activate a pathway which would otherwise only be activated if a somatic mutation would have occurred thereby mimicking the effect of a driver mutation for cancer.

Project description:Our findings establish a key role for the coregulator, Repressor of Estrogen receptor Activity (REA), in controlling the timing and magnitude of decidualization in human endometrial stromal cells in vitro and in the mouse uterus in vivo, and suggest that REA functions to synchronize uterine differentiation with concurrent embryo development, which is essential for optimal implantation and fertility. The findings highlight that REA physiologically restrains endometrial stromal cell decidualization, controlling the timing and magnitude of decidualization to enable proper synchronization of uterine differentiation with concurrent embryo development that is essential for implantation and optimal fertility.

Project description:Natural killer (NK) cell function is enhanced by interaction of inhibitory NK receptors with MHC class I molecules. NK cell function is critical during placentation, where these cells mediate adaptations to pregnancy in concert with other cells at the maternal/fetal interface. The aim of this experiment was to determine the downstream consequences on the uterine transcriptome that arise from lack of NK interaction with MHC class I molecules. This was done in mice lacking beta-2-microglobulin, thus not having correctly folded surface MHC class I. Biopsies were obtained from the mesometrial poles of the implantation chamber in gestation day (gd) 10.5 pregnant mice.

Project description:Uterine injury from procedures such as Cesarean sections (C-sections) often have severe consequences on subsequent pregnancy outcomes, leading to disorders such as placenta previa, placenta accreta, and infertility. With rates of C-section at approximately 30% of deliveries in the US that are projected to continue to climb, an understanding of the mechanisms by which these pregnancy disorders arise and opportunities for intervention are sorely needed. However, there are currently no animal models to date that comprehensively assess the consequences of uterine injury. Here we describe the first rodent model of uterine injury on subsequent in utero outcomes. We observed four distinct phenotypes: reduced implantation events, increased rates of resorption, embryo misspacing, and placenta accreta-like features of reduced decidua and expansion of invasive trophoblasts. We also show that the appearance of embryo misspacing depends entirely on the phase of estrous cycle at the time of injury, leading us to identify differences in uterine wound healing as a function of estrous. Using RNA-seq, we identified perturbations in the expression of components of the COX-2 pathway after recovery from injury, a pathway that has previously been demonstrated to play an important role in embryo spacing. Therefore, we demonstrate that uterine damage in our novel mouse model engenders many uterine changes, most notably perturbed expression of COX-2 pathway-related genes, that ultimately lead to numerous placental and embryonic developmental defects with parallels to human pregnancy disorders.
