Project description:Whole genome bisulfite sequencing (WGBS‑seq) and whole-genome oxidative bisulfite sequencing (oxWGBS‑seq) in four paired OSCC tumor and its NAT were performed to obtain precise, genome-wide, single-base resolution maps of 5mC and 5hmC. Based on these maps, the genomic content of 5mC and 5hmC in these paired samples were observed, as well as the distributions of 5mC and 5hmC in genic regions and histone-marked gene regulatory regions.

Project description:Chemosensitization is an effective strategy to overcome the drawbacks of arsenic trioxide (As2O3) treatment which may be possible through the use of dietary supplements in combination. Proteomic analysis revealed that the combination of arsenic trioxide with vitamins can alter a number of proteins related to the translation, cell cycle, oxidative stress and apoptotic pathways that adds to their effectiveness in the treatment of acute promyelocytic leukemia. This study provides the fact that HL-60 cells became more vulnerable to As2O3 in the presence of L-Ascorbic acid and α-Tocopherol indicating that this combination may be a hopeful approach to increase the outcome of As2O3 in the treatment of acute promyelocytic leukemia.
Project description:In this study, we show that by simple modulation of extrinsic signaling pathways, a new class of pluripotent stem cells, referred to as region selective epiblast stem cells (rsEpiSCs), could be efficiently derived from different stages of the early embryo. rsEpiSCs share features of primed pluripotency yet are distinct from EpiSCs in their molecular characteristics and ability to colonize post-implantation embryos. We performed whole-genome bisulfite sequencing (WGBS) experiments to compare the DNA methylation landscapes of conventional EpiSCs and rsEpiSCs. Compare the DNA methylation profiles in 2 pluripotent stem cell types (LP-EpiSCs and conventional EpiSCs) in mouse. Two replicates are examined for each cell type.

Project description:We wanted to emphisize the role of intragenic CpG island in various cell differentiation. We especially examined the effect of DNA methyltransferase (DNMT) in the neural differentiation from mouse ES cell. Therefore, we performed the Whole-Genome Bisulfite Sequencing (WGBS) analysis of mouse ES cell, ESC-derived NPC, and also in DNMT double knockout (DKO) conditions. By utilizing these result, we confirmed the genes regulated by intragenic CpG island methylation have effect in NPC differentation.
