Transcriptome analysis of endometrial organoids derived from patients with Mayer-Rokitansky-KÃƒÂ¼ster-Hauser syndrome

ABSTRACT: The Mayer-Rokitansky-KÃƒÂ¼ster-Hauser (MRKH) syndrome is characterized by agenesis of the uterus and upper part of the vagina in females with normal ovarian function. The cause of the disease is still enigmatic. Here, we developed organoid cultures from endometrium found in uterine rudiment horns of MRKH patients. Phenotypically they share great similarity between healthy control organoids and are fully hormone responsive. Transcriptome analysis using RNA-seq identified possible disease-causing pathways altered in MRKH patients during development of the female reproductive tract. Thus, the organoid cultures provide a powerful research model for further insight into disease-causing alterations.

The endometrial transcription landscape of MRKH syndrome

Project description:The Mayer-Rokitansky-KÃƒÂ¼ster-Hauser (MRKH) syndrome (OMIM 277000) is characterized by agenesis of the uterus and upper part of the vagina in females with normal ovarian function. While genetic causes have been identified for a small subset of patients and epigenetic mechanisms presumably contribute to the pathogenic unfolding, too, the etiology of the syndrome has remained largely enigmatic. A comprehensive understanding of gene activity in the context of the disease is crucial to identify etiological components and their potential interplay. So far, this understanding is lacking, primarily due to the scarcity of samples and suitable tissue. In order to close this gap, we profiled endometrial tissue of uterus rudiments in a large cohort of MRKH patients using RNA-seq and thereby provide a genome-wide view on the altered transcription landscape of the MRKH syndrome.

Apelin-13 modulates the endometrial transcriptome of the domestic pig during the implantation phase of pregnancy

Project description:Apelin is a hormone secreted primarily by white adipose tissue. Recent studies have shown that adipokine and its receptor, APJ, are expressed in different reproductive tissues, including the ovary and uterus, which suggests that it may act as an important local regulator of reproductive functions. This study aimed to explore the influence of apelin on global gene expression in the porcine endometrium during early gestation, on days 15 to 16 of pregnancy (implantation period). We analysed samples obtained from 4 individuals (n=4). The tissue explants obtained from the uterine horns were divided into controls [without treatment] and apelin-13 (20 ng/mL) treated ones. In vitro tissue cultures were conducted for 24 hours.

Menstrual flow as a non-invasive source of endometrial organoids

Project description:Organoid cultures derived from menstrual flow faithfully replicate secretory phase endometrial glands and provide a novel, non-invasive technique for the clinical assessment of endometrial function. Paired organoid cultures derived from scratch biopsies and ensuing menstrual flow share the same transcriptome signature, responses to early pregnancy hormones, and pathological changes in cases of early-pregnancy loss. The technique opens new avenues for investigating endometrial function in cases of subfertility and gynaecological disorders.

Gene expression signature of endometrial samples from women with and without endometriosis.

Project description:Endometriosis is a benign gynecological disease characterized by the presence of endometrial-like cells outside the uterus. The prevalence of this disease reaches 10-15% in women of reproductive age which accounts for approximately 200 mln of women worldwide. The existing therapeutic approaches include medications and surgical options, but the risk of post-treatment recurrence of endometriosis remains high. There are currently no effective biomarkers of endometriosis in clinical practice and laparoscopy remains the gold standard for its diagnosis. However, laparoscopy is highly invasive procedure that frequently leads to complications and cannot be used as a routine screening test. In this study we profiled by RNA sequencing 52 endometrial pathological samples and 12 corresponding normal human tissues. We performed two-step differential gene expression analysis of endometrial samples and lesions of patients with endometriosis in comparison with normal reproductive tissues. Based on this analysis we generated a characteristic signature of five genes downregulated in endometriosis with respect to healthy endometrium. The 5-gene expression signature identified had significant predictive power (AUC>0.85) thus suggesting that the marker gene set identified can be used for a low invasive molecular diagnostic of endometriosis. Our data also suggest that the statistical method of five-fold cross validation of differential gene expression analysis procedure can be used for the ability to generate robust gene signatures using real-world clinical data.

Self-renewing Endometrial Epithelial Organoids of the Human Uterus [bulk RNA-seq data]

Project description:The inner lining of the human uterus, termed the endometrium, is replete with glands that are hypothesized to be important for establishment of pregnancy through interactions with other endometrial cells and the embryo. A three-dimensional culture system was utilized to establish and generate endometrial epithelial organoids from normal human endometrium that display long-term expandability, can be cryopreserved, and are hormone responsive. Here, single cell RNA-seq was used to create a high-resolution gene expression atlas of the endometrial epithelial organoids and to define responsiveness to reproductive hormones.

A multiplexing strategy to create primary and orthotopic endometrial cancer models in mice

Project description:Endometrial cancer (EC) is the most common female reproductive tract cancer and its incidence is continuously rising recently. The underlying mechanisms of EC tumorigenesis remain unclear and efficient target therapies are lack, both of which require feasible endometrial cancer animal models for translational studies. Here, we report an organoid and genome editing-based strategy to generate primary, orthotopic, and drivers-defined ECs in mice. Bulk RNA sequencing was performed with organoids of various genetics-mediated EC models.

TGFβ signaling in the uterine epithelium controls endometrial cell homeostasis and regeneration [RNA-Seq]

Project description:In this study, we define the epithelial-specific contribution of SMAD2/3 to endometrial function by conditionally ablating the Smad2 and Smad3 transcription factors using Lactoferrin-iCre (Ltf-cre). Using epithelial organoid cultures from the endometrium, we uncover the signaling mechanisms downstream of TGFβ that control endometrial cell regeneration and are critical for endometrial regeneration and homeostasis.
