ABSTRACT: This project aims to characterize Italian maize landraces both morphologically and genotypically to assess their genetic diversity and identify climate-adaptive alleles for future pre-breeding initiatives. To achieve this, we sequenced 140 individuals from 28 representative Italian maize landraces from northern and central Italy using the Genotyping-by-Sequencing (GBS) approach

Mexican maize landraces under drought stress and recovery irrigation

Project description:Transcriptional profiling of three mexican maize landraces under 10, and 17 days stress and recovery irrigation A dye balanced modified loop design was implemented. Two biological replicates (pooling five representative plants) representing each sampling point for each genotype were obtained for purified RNA from 120 randomly chosen seedlings. This experiment involved a total of forty-eight (24 sets) of microarray hybridizations, including direct and dye swap comparisons between treatments as well as across the three landraces. This design allowed us to determine differences in gene expression between the three different landraces under drought stress (10 and 17 days) and at recovery irrigation compared to irrigated controls.

W23 and High altitude maize under solar and supplementary UV-B

Project description:Additional to GSE1807 A comparative analysis, by expression profiling of maize, was performed to identify novel components in the mechanisms of maize responses to UV-B. Five high-altitude landraces grown from 2,000 to 3,400 m naturally receive higher UV-B fluence than plants at lower altitudes and similar latitudes. These high-altitude landraces were compared directly with a low-altitude line and with literature reports for other temperate maize lines. A microarray analysis demonstrated that among the UV-B responsive transcripts, several types of gene implicated in chromatin remodeling are differentially expressed before and after UV-B treatment in high-altitude lines. RNAi transgenic plants with lower expression of four such chromatin-associated genes exhibited hypersensitivity to UV-B by measurements of leaf arching, increased leaf chlorosis and necrosis, and altered UV-B regulation of selected genes. These results collectively suggest that genes involved in chromatin remodeling are crucial for UV-B acclimation and that some high-altitude lines exhibit adaptations to this challenge.

Gene expression of maize-specific Exserohilum turcicum strain

Project description:Exserohilum turcicum is a fungal pathogen that causes northern corn leaf blight and sorghum leaf blight. However, strains are host-specific, meaning a strain typically only causes disease on either maize or sorghum. This pathosystem provides a unique opportunity to identify the mechanisms underlying host specificity through genomic and transcriptomic studies. To characterize host specificity, we (i) constructed de novo annotated assemblies of one sorghum- (15St008) and one maize-specific (Et52B) E. turcicum strain, ii) identified and characterized structural changes between the two genomes, and (iii) compared differential gene expression. This GEO project contains the gene expression of the maize-specific strain (Et52B) during in planta infection of maize and in axenic cultures.

The Exserohilum turcicum effector EtEC81 reprograms alternative splicing in maize and activates immunity [RIP-Seq]

Project description:Some pathogen-derived effectors reprogram mRNA splicing in their host plant to regulate plant immune responses. The fungus Exserohilum turcicum is the causal agent of northern corn leaf blight, a damaging maize (Zea mays) disease. However, the low efficiency of genetic transformation of E. turcicum has hampered research on its effectors and whether E. turcicum effectors interfere with RNA splicing remained unknown. Here, using an alternative splicing (AS) reporter system, we identified the secreted protein EtEC81 (Exserohilum turcicum effector 81), which modulates the AS of maize pre-mRNAs and negatively regulates the pathogenicity of E. turcicum. EtEC81 physically interacts with EtEC81-interactiNG protein 1 (ZmEIP1), which associates with maize spliceosome components, regulating AS and positively regulating the defense response against E. turcicum. EtEC81 binding further enhanced the effect of ZmEIP1 on AS. Transcriptome analysis revealed 119 common events with altered AS in maize plants transiently overexpressing ZmEIP1 or EtEC81, suggesting that these factors cause the mis-regulation of cellular activities and thus induce immune responses. We used RT-qPCR to verify representative AS events in the plants transiently overexpressing ZmEIP1 and EtEC81. Together, our results suggest that the EtEC81 effector targets ZmEIP1 to reprogram pre-mRNA splicing in maize.
