Project description:DNA methylation patterns reflect the status of individual tissues, such as cell composition, age, and the local environment in mammals. This experiment addressed the DNA methylation landscape in the dog genome across three breeds: Shiba, Dachshund (Miniature), and Poodle (Toy). A comprehensive profile of whole-genome DNA methylation from the whole blood of three dog breeds was generated using whole-genome bisulfite sequencing.

Determination of animal species origin of blood in artworks by LC-MS/MS

Project description:For art historians, understanding the animal origin of blood used in artworks is crucial for interpreting materials, techniques, and historical practices. It provides insight into the cultural, and religious significance of the work and allows for a more nuanced interpretation of its symbolic meaning. In this work, sets of peptides of blood proteins, more specifically their amino acid sequences, that are responsible for the differences between seven animal species (cat, cow, dog, goose, hen, human, and pig) were identified by LC-MS/MS. Protein materials used for the preparation of six model blood coatings were confirmed and the animal origin of the used blood was found. Finally, the blood origin in four samples taken from a set of Japanese and Chinese lacquer artworks from 18th and 19th century was also determined.

Delayed protein translocation protects mitochondria against toxic CAT-tailed proteins

Project description:In the ribosome-associated quality control acting on nuclear-encoded mitochondrial proteins (mitoRQC), Vms1 protects mitochondria from toxic effects of Rqc2-generated C-terminal alanyl and threonyl (CAT) tailed proteins that escaped proteosomal degradation facilitated by E3 ubiquitin ligase Ltn1. Here, we performed a genome-wide screen in yeast to identify novel proteins involved in mitoRQC. We found that Pth2, a peptidyl-tRNA hydrolase in the outer mitochondrial membrane, influences aggregation of mitochondrial-targeted CAT-tailed proteins without majorly affecting the CAT-tailing process itself. Peptidyl-tRNA hydrolase activity is essential during this process, however, the activity of Pth2 can be substituted by another peptidyl-tRNA hydrolase, upon proper localization. Our data suggest that Pth2 acts through modulating protein import into mitochondria, enabling CAT-tailed proteins to get access to the cytosolic chaperones and thus relieving the mitochondrial proteostasis network. Other hits obtained in the screen show that, in general, slowing down protein translocation protects mitochondria against toxic CAT-tailed proteins.
