SNP panel development for genetic management of wild and domesticated white bass (Morone chrysops)

ABSTRACT: In this study, we developed SNP resources to examine the genetic relationships among a long-term domesticated white bass line and five potential founder stocks. A final set of 57- SNP assays was successfully designed and validated using MassARRAY system. The SNP resources developed in this study should facilitate ongoing efforts in selective breeding and conservation of white bass.

Project description:The aim of this study was to identify genes expressed selectively in brown adipose tissue as compared to white adipose tissue from the same animals. This analysis provides a gene set that is brown and white adipose selective. Experiment Overall Design: Interscapular brown adipose tissue and epididymal white adipose tissue was carefully dissected from 3 male C57Bl/6 mice. These samples were profiled independently using Affymetrix mouse 430_2 gene arrays, representing 3 biological replicates for each brown and white adipose tissues.

Transcriptome analysis of adductor muscle color in Boihai Red Scallop

Project description:Boihai Red is new strains of inter-specific hybridizing the bay scallop (Argopecten irradians irradians) with the Peruvian scallop (Argopecten purpuratus). Orange color variant of adductor muscle have been developed through successive selective breeding in this strain. In the present study, transcriptomic was conducted on orange and white adductor muscle tissues. Transcriptomic analysis showeds 416 differentially expressed genes (DEGs) were identified between white and orange adductor muscle tissues in Boihai Red Scallop, with 216 up regulated and 200 down. In DEGs, apolipophorin, CYP450 and tyrosinase were expressed highly in orange adductor muscle tissues, which related to carotenoids or melanin. It is probable that not only carotenoids, but also melanin act on orange color of adductor muscle. This study provides valuable genetic resources for understanding underlying mechanisms and pathways of adductor muscle color.

Project description:Purpose: Determine the function of lncBATE10 in brown and white adipocyte differentiation Methods: primary brown and white apreadipocytes were isolated by cell culture. We infected brown preadipocytes with retroviral shRNA targeting lncBATE10 or with retrovirus overexpressing lncBATE10. Cells were induced to differetinate for 5 days. Total RNA were harvested for RNA-seq Conclusions: Our study shows that lncBATE10 is required for BAT-selective program expression. Overexpression of lncBATE10 is not sufficient to promote BAT marker expression.

JPST000091-2 White adipocyte phosphoproteomics [Reanalysis: JPST000091]

Project description:Catecholamines promote lipolysis both in brown and white adipocytes, whereas the same stimuli preferentially activate thermogenesis in brown adipocytes. Molecular mechanisms for the adipose-selective activation of thermogenesis remain poorly understood. Here, we employed quantitative phosphoproteomics to map global and temporal phosphorylation profiles in brown, beige, and white adipocytes under Î²3-adrenenoceptor activation and identified kinases responsible for the adipose-selective phosphorylation profiles. We found that casein kinase2 (CK2) activity is preferentially higher in white adipocytes than brown/beige adipocytes. Genetic or pharmacological blockade of CK2 in white adipocytes activates the thermogenic program in response to cAMP stimuli. Such activation is largely through reduced CK2-mediated phosphorylation of class I HDACs. Notably, inhibition of CK2 promotes beige adipocyte biogenesis and leads to an increase in whole-body energy expenditure and ameliorates diet-induced obesity and insulin resistance. These results indicate that CK2 is a plausible target to rewire the Î²3-adrenenoceptor signaling cascade that promotes thermogenesis in adipocytes. [Original project description]
