Cloning of wheat leaf rust resistance gene LrLC10

ABSTRACT: Wheat leaf rust resistance gene LrLC10 is highly effective to leaf rust, we re-sequenced the parental lines (Liaochun10 and 7D49) to discover the variations (SNPs and indel) on the candidate region of LrLC10 and convert them to markers to fine map the gene. It started in 2017 and is based at wheat genetic and genome center.

Small RNA, transcriptome and degradome analysis of the transgenerational heat stress response network in durum wheat

Project description:Heat stress is a major limiting factor for grain yield and grain quality in wheat production. In crops, abiotic stresses have transgenerational effects and the mechanistic basis of stress memory is associated with epigenetic regulation. The current study presents the first systematic analysis of the transgenerational effects of post-anthesis heat stress in tetraploid wheat. Genotype-dependent response patterns to parental and progeny heat stress were found for the leaf physiological traits, harvest components, and grain quality traits measured. Parental heat stress had positive influence on the offspring under re-occurring stress for traits like chlorophyll content, grain weight, grain number and grain total starch content. Integrated sequencing analysis of the small RNAome, mRNA transcriptome, and mRNA degradome provided the first description of the molecular networks mediating heat stress adaption under transgenerational influence. The expression profile of 1771 microRNAs (733 being novel) and 66,559 genes was provided, with differentially expressed microRNAs and genes identified subject to the progeny treatment, parental treatment and tissue type factors. Gene Ontology and KEGG pathway annotation of stress responsive microRNAs-mRNA modules provided further information on their functional roles in biological processes like hormone homeostasis, signal transduction, and protein stabilization. Our results provide new sights on the molecular basis of transgenerational heat stress adaptation, which can be used for improving thermos-tolerance in breeding.

Expression data from rust or mock inoculated, fully expanded flag leaf halves

Project description:Two sets of wheat lines near-isogenic to Lr34 were used to compare gene expression profiles of wheat: 1. with and without Lr34 gene; 2. rust and mock inoculation; 3. distal and basal portion of the flag leaves. The two sets of wheat near-isogenic lines were used to subtract genetic background variations and to enrich Lr34-regulated gene expression profiles. The study is aimed to better understand the mechanisms of the well-known durable leaf rust resistance gene, Lr34, mediated resistance at the transcriptome level. Global transgenerational gene expression dynamics in two nascent allohexaploid wheat lines analogous in genome constitution to common wheat (Triticum aestivum)

Project description:Alteration in gene expression accompanying initial stages of allopolyploidy is a prominent feature in plants, but its spectrum and model are highly idiosyncratic. We used multi-colour GISH to identify individuals from two nascent allohexaploid wheat lines between Triticum turgidum and Aegilops tauschii, which had a transgenerationally stable chromosomal constitution mimicking that of common wheat. We performed genomewide analysis of gene expression for these plants along with their parental species using the Affymetrix GeneChip Wheat Genome-Array. Comparison with parental species coupled with inclusion of empirical mid-parent values (MPVs) revealed two patterns of alteration in gene expression in the allohexaploid lines: parental dominance expression and nonadditive expression. Genes involved in each altered pattern could be classified into three distinct groups, stochastic, heritable and persistent, based on their transgenerational heritability and inter-line conservation. Whereas both altered patterns of gene expression showed a propensity of inheritance, identity of the involved genes is stochastic, consistent with the involvement of diverse Gene Ontology (GO) terms. Nonetheless, those genes showing nonadditive expression exhibited a significant enrichment for vesicle-function. Our results suggest global alteration in gene expression conditioned by nascent allopolyploidy likely play functional roles in stabilization and establishment of the newly formed plants, and consequential to evolution. We used Affymetrix GeneChip Wheat Genome-Array to compare the expression difference of newly synthetic wheat allopolyploid lines and their parental lines.

W4909 and W4910 are two lines derived from crossing AJDAj5 and PhI, and both are more salt tolerant than either parental line.

RIL89 is derived from a cross between parental lines B73 and H99.

Transcript profiling of Lr1- and Lr34-mediated leaf rust resistance in wheat

Project description:The wheat gene Lr34 confers partial resistance to all races of Puccinia triticina, the causal agent of wheat leaf rust. However, the biological basis for the exceptional durability of Lr34 is unclear. The Affymetrix wheat genome array was used to identify wheat genes differentially expressed in a compatible interaction (Tc), an R-gene mediated incompatible interaction (Tc-Lr1), and a race non-specific resistance interaction (Tc-Lr34) in response to infection challenge by P. triticina race 1 at anthesis. Identification of Candidate Master Regulators of the Response to Early Heat Stress in Climate-adapted Wheat Landraces via Transcriptomic and Co-expression Network Analyses

Project description:Climate change is not only causing a rise of global temperature but also more variable climates. In major wheat-producing countries, spring temperatures have increased up to 40°C in recent years. Considering the wheat optimal growth temperature of around 20°C, wheat is particularly prone to damage by heat stress. To combat this threat, it is imperative to understand the response of wheat to early heat stress. This research is focus on understanging the transcriptional reprogramming of wheat when exposed to heat stress during the three-leaf stage. Differential expression and weighted gene co-expression network analysis have been used here before and after the stress to to identify candidate master-regulators of this transcriptional response.
