BAFF SNPs and fatigue in primary SS patients

ABSTRACT: An investigation into the association of BAFF variants with fatigue development in the setting of primary Sjogren's Syndrome (SS). Analysis performed in a cohort of 199 Greek primary SS patients, in a validation cohort of 62 Dutch primary SS patients and in a cohort of 52 Greek Multiple Sclerosis (MS) patients that served as disease controls.

Project description:The pathogenesis of multiple sclerosis (MS) remains to be elucidated. Pediatric-onset MS (POMS) represents the earliest stage of the disease. CSF proteins in POMS may therefore provide causal information. Therefore, the aim of the current study was to analyze CSF proteins in children with an initial CNS acquired demyelinating syndrome (ADS) and to make a comparison between POMS and monophasic ADS (mADS). Patients were selected from two prospective pediatric ADS studies. Liquid chromatography-mass spectrometry was performed in a Dutch discovery cohort (POMS n=28; mADS n=39). Parallel reaction monitoring-mass spectrometry was performed on selected proteins more abundant in POMS with ≥ 8 unique peptides in a combined Dutch and Canadian validation cohort (POMS n=48; mADS n=106).

Cohort 2: miRNA expression profiling in the whole blood of Primary Sjogren's Syndome patients

Project description:Whole blood total RNA was isolated form PaxGene tubes using the PaxGene total RNA extraction kits. miRNA expression was profiled in Primary Sjogren's Syndrome patients and controls in order to independently validate differentially expressed miRNAs between patients and controls and between patient subgroups that were identified in 'Cohort 1'.

Proteomic analysis of sweat samples from tuberculosis patients

Project description:We performed a global proteomic profile of eccrine sweat sampled from patients with active pulmonary TB, other lung diseases (non-TB disease), and healthy controls. A comparison of proteomics results between Active-TB, Non-TB and Healthy Controls was done in search for proteins unique to the active TB-patient cohort as well as proteins unique to those without TB disease. We conclude tthat global proteomic profiling of eccrine sweat collected from TB and non-TB patients is a viable approach for the identification of unique proteins differentially present based on clinical cohort. These proteins may represent unique biomarker profiles that upon further verification and validation could be used to develop a non-sputum based test for diagnosis of active TB.

Project description:Purpose: The goal of this experiment was to identify differentially expressed miRNAs between colon cancer patients with and without metachronous metastases. Background: Colon cancer prognosis and treatment are currently based on a classification system still showing large heterogeneity in clinical outcome, especially in TNM-stages II-III. Prognostic biomarkers for metastasis risk are warranted, as development of distant recurrent disease mainly accounts for the high lethality rates of colon cancer. MicroRNAs have been proposed as potential biomarkers for cancer. Furthermore, a verified standard for normalization of the amount of input material in PCR-based relative quantification of miRNA expression is lacking. Methods: A selection of frozen tumor specimens from two independent patient cohorts with TNM-stage II-III microsatellite stable primary adenocarcinomas were used for laser capture microdissection. Next-generation sequencing was performed on small RNAs isolated from colorectal tumors from the Dutch cohort (N=50). Differential expression analysis, comparing in metastasized- and non-metastasized tumors, identified prognostic microRNAs. Validation was performed on colon tumors from the German cohort (N=43) using qPCR. Results: MiR-25-3p and miR-339-5p were identified and validated as independent prognostic markers and used to construct a multivariate nomogram for metastasis risk prediction. The nomogram showed good probability prediction in validation. Additionally, we recommend combination of miR-16-5p and miR-26a-5p as standard for normalization in qPCR of colon cancer tissue-derived microRNA expression. Conclusions: In this international study, we identified and validated an miRNA classifier in primary cancers, and propose a nomogram capable of predicting metastasis risk in microsatellite stable TNM-stage II-III colon cancer.
