Project description:We conducted prospective clinical studies to validate the importance of CD4+ T cells in 13 diseases from the following ICD-10-CM chapters: Neoplasms (breast cancer, chronic lymphocytic leukemia); endocrine, nutritional and metabolic diseases (type I diabetes, obesity); diseases of the circulatory system (atherosclerosis); diseases of the respiratory system (acute tonsillitis, influenza, seasonal allergic rhinitis, asthma); diseases of the digestive system (Crohn’s disease [CD], ulcerative colitis [UC]); and diseases of the skin and subcutaneous tissue (atopic eczema, psoriatic diseases). Study participants were recruited by clinical specialists based on diagnostic criteria defined by organizations representing each specialist’s discipline. Age and gender matched healthy controls (n = 127 and 39, respectively) were recruited in the Southeast region of Sweden from outpatient clinics at the University Hospital, Linköping; Ryhov County Hospital, Jönköping, a primary health care center in Jönköping; and a medical specialist unit for children in Värnamo. Study participants represented both urban and rural populations with an age range of 8–94 years. Patients with type I diabetes and obesity had an age range of 8–18 years. 12 patients had more than one diagnosis.

Project description:With increasing age of the global population, age-associated diseases such as cardiovascular diseases, cancer or diabetes are on the rise in many countries. To identify cellular processes and markers of aging in humans, this study performed comparative whole genome microarray gene expression profiling of peripheral blood mononuclear cells (PBMC) isolated from two age groups, i.e., an old age group (age: 75-89 years) and a younger, middle-aged group (age: 35-50 years). Total RNA was isolated from the PBMC and subjected to whole genome microarray gene expression profiling. The whole genome gene expression data of human PBMC reveal age-associated changes in transcript levels and contribute to a better understanding of biological processes underlying aging and the pathogenesis of age-associated diseases.

Project description:Obesity is a chronic inflammatory condition which leads to a change in the production of adipokines and cytokines that can be found in blood plasma and saliva. It increases the risk for periodontal and cardiovascular diseases, hypertension, dyslipidemia, type 2 Diabetes Mellitus, cancer and others. This study aims to analyze the plasma and saliva proteomes of young adults with obesity and overweight comparing to normal weight individuals, to reveal the influence of body index in the proteome profiles and to identify early biomarkers of obesity-associated diseases.

Project description:Background: The obesity epidemic and the aging population of many western countries together with their associated diseases are major challenges for the healthcare. As obesity is a risk factor for many age related diseases, such as cancer, it is of importance to understand their interaction and the underlying molecular mechanisms. Lately, epigenetic programming in the form of DNA methylation and have been recognized for its importance in aging, obesity and several diseases. Method: Herein, the methylation level was determined for around 27000 CpG-islands in 46 DNA samples from adult peripheral blood with microarrays. The dependence for each CpG island with age, obesity and their interaction was ascertained with a general linear model. Results: The methylation level of more than 100 genomic sites were significantly altered with increasing age together with 10 additional sites that were differentially methylated with age in obese and lean individuals. The majority of the genomic sites were hypermethylated during aging, including the telomerase catalytic subunit (TERT). However, age dependent hypermethylation was prohibited or reverted in 8 of 10 regions in obese where an interaction between age and obesity was observed. Moreover, one region (LINC00304) was differentially methylated in obese and lean. Conclusion: This study provides evidence for an obesity influence on age driven epigenetic changes, which provide a molecular link between aging and obesity. This link and the identified markers may prove to be valuable biomarkers for the understanding of the molecular basis of aging, obesity and associated diseases. Project description:Obesity is a growing epidemic worldwide and is a major risk factor for several chronic diseases, including diabetes, kidney disease, heart disease, and cancer. The global epidemic of obesity has been accompanied with a dramatic increase in the incidence and prevalence of type 2 diabetes mellitus (T2DM). Obesity often leads to T2D, via the increased production of proinflammatory cytokines such as tumor necrosis factor alpha. Our study combines different proteomic techniques to investigate the changes in the global proteome, secretome and phosphoproteome of adipocytes under chronic inflammation condition, as well as fundamental cross-talks between different cellular pathways regulated by chronic TNF alpha exposure. Our results show that many key regulator proteins of the canonical and non-canonical NF-kB pathways, such as Nfkb2, and its downstream effectors, including Csf-1 and Lgals3bp, directly involved in leukocyte migration and invasion, were significantly upregulated at the intra and extracellular levels, culminating in the progression of inflammation. Our data provides evidence of several key proteins that play a role in the development of insulin resistance.

Project description:Chronic Kidney Disease (CKD), a global health burden, is strongly associated with age-related renal function decline, hypertension and diabetes, frequent consequences of obesity. Despite extensive studies the mechanisms determining susceptibility to CKD remain elusive. Clinical evidence together with prior studies from our group showed that perinatal metabolic disorders after maternal obesity adversely affect kidney structure and function throughout life. Since obesity and aging processes converge in similar pathways we tested if perinatal obesity induced by High-Fat Diet (HFD)-fed female mice sensitizes aging-associated mechanism in kidneys of newborn mice. Tissue from the kidney cortex and the kidney medulla was collected from offspring of control or HFD-fed mice.

Project description:Obesity represents a global burden with an increasing worldwide prevalence, especially in women of reproductive age. Obesity in women, defined as a body mass index (BMI) > 30 kg/m2, has a worldwide prevalence ~21%, however, it exceeds 30% in countries such as Chile, Mexico, United States, and the United Kingdom. Growing evidence support the notion that pre-gestational obesity confers an increased risk for the development of diabetes, obesity and chronic inflammatory diseases in the offspring later in life. In fact, maternal obesity during gestation is associated with an increased risk of asthma and wheezing during early or late infancy, in terms of medication and need for hospital admission. Here we report changes in DNA methylation at a genomic level in monocytes isolated by adherence from cord blood of neonates born to women with obesity and integrate these changes to reveal the epigenetic programming of immune signaling pathways resulting from maternal obesity.
