Integrative Genomic Analysis Reveals Shared Loci for Reproductive and Productive Traits in Yorkshire Pigs

ABSTRACT: This project focuses on the integrative genomic analysis of Yorkshire pigs, aiming to identify shared loci associated with both reproductive and productive traits. By leveraging genomic data, we seek to uncover genetic factors that influence key characteristics in this breed, enhancing breeding strategies and improving overall productivity. The findings from this study are expected to contribute valuable insights into the genetic basis of traits critical for livestock improvement.

Project description:Reproductive efficiency in sheep is critically important for livestock production. This study provides the first multi-tissue proteomic characterization of high fecundity in Hu sheep, suggesting that their reproductive superiority may originate from hypothalamic-driven regulation, efficient coordination of key signaling pathways such as cAMP, and complex interactions among functionally important proteins across tissues. These findings offer valuable molecular insights into the regulation of reproductive traits in sheep, and the identified key proteins and pathways represent promising targets for further functional validation and molecular breeding applications.

Project description:The clinical and pathological features of early-onset colorectal cancer (EOCRC) differ from those of late-onset colorectal cancer (LOCRC). Our research aims to thoroughly elucidate the distinctions between them by analyzing clinical prognosis, metastatic patterns, gene expression, and genomic mutation profiles. Our deliberation will uncover latent strategies for personalized therapeutic of both EOCRC and LOCRC.

mRNA-seq of individual genetically identical Caenorhabditis elegans adults

Project description:Individual phenotypic differences persist even in genetically identical individuals, although separating genetic and environmental causation is difficult or impossible in most organisms. To understand the basis of individual differences in the absence of genetic differences, we measured two quantitative reproductive traits in genetically identical young adult Caenorhabditis elegans roundworms in a shared environment and performed single-individual mRNA-seq on each worm. We identified hundreds of genes for which expression variation was strongly associated with reproductive traits, some of which depended on prior environmental experience and some of which was random. Multiple small sets of genes together were highly predictive of reproductive traits across individuals.

Multimodal stimulation screens reveal unique and shared regulators limiting T-cell fitness [Ctbp1]

Project description:It has not been systematically studied whether T-cell fitness regulators uniquely or broadly contribute to specific traits limiting antitumor activity. We performed genome-scale CRISPR/Cas9 knockout screens in primary CD8 T-cells to uncover genes negatively impacting on their fitness upon three modes of stimulation: (1) intense stimulation, causing activation-induced cell death (AICD); (2) acute stimulation, triggering T-cell expansion; (3) chronic stimulation, resulting in dysfunction. Besides established regulators, we uncovered several genes controlling T-cell fitness either specifically or commonly under various stimulation modalities. Ablation of Dap5, ranking highly in all three screens, increased translation while enhancing tumor killing. Loss of Icam1-mediated homotypic T-cell clustering amplified T-cell expansion and effector functions after both acute and intense stimulation. Finally, inactivation of Ctbp1 induced T-cell persistence exclusively upon chronic stimulation. Our results functionally annotate T-cell fitness regulators based on their unique or shared contribution to traits limiting T-cell antitumor activity.
