Project description:The defining characteristics of leukemia, such as lineage and genetics, are associated with a typical age of onset. Understanding mechanisms of leukemia age specificity could improve disease models to develop new therapies. We used heterochronic transplantation of murine leukemia driven by MLL-AF9 to investigate the relative contribution of the age of the cell-of-origin or the hematopoietic microenvironment to the lineage fate of leukemia initiating cells (LICs). We show that the neonatal hematopoietic niche supports the development of infant-like mixed lineage B-cell/myeloid leukemia, while a mature niche promotes adult-like pure acute myeloid leukemia (AML) from identical cells of origin. We attribute this to inhibition of B-lymphoid fate in multipotent progenitor-like LICs by Ccl5 from adult bone marrow (BM) stroma, and implicate glycogen synthase kinase-3 (GSK-3) signaling in the myeloid fate specification in mouse and human MLL-driven leukemia. These findings connect maturation and aging of the hematopoietic system to leukemia age specificity.

Leveraging these advantageous characteristics requires an in-depth understanding of parameters that have shaped sequence variation in isolates. Unearthing Prehistoric Diets: First Evidence of Horse Meat Consumption in Early Bronze Age Sicily

Project description:This paper presents the earliest documented evidence for the presence and consumption of horse meat in Early Bronze Age Sicily, significantly revising previous understandings of equid use on the island. Multidisciplinary analyses involving proteomics and lipidomics were performed on ceramic vessels from the Castelluccian settlement at Polizzello Mountain (Caltanissetta), revealing residues consistent with equine-derived substances. Proteomic data unequivocally identified equine serum albumin in multiple pottery fragments, demonstrating active consumption or processing of horse-derived substances within a ceremonial or dietary context. Lipid residues further supported this interpretation, indicating the presence of animal fats and vegetable-derived substances within the pottery. These findings substantially alter existing models of horse domestication, utilization, and dietary practices in prehistoric Sicily, suggesting a far earlier and more complex human-equid relationship. Furthermore, the integration of biomolecular data enhances our understanding of intercultural interactions, ritual behaviors, and economic strategies in the central Mediterranean during the third millennium BCE.

Dairy pastoralism sustained Eastern Eurasian steppe populations for 5000 years

Project description:Through the use of proteomic analysis of 32 dental calculus samples from anceint Mongolia, we show that ruminant dairying was present in Mongolia by at least 3000 B.C.E., over 1500 years prior to previously published work. Excitingly, the earliest site with dairy evidence has been identified as archaeologically Afanasievo, supporting the hypothesis that dairy practices and animals likely entered the Eastern steppe with migrating western steppe populations. Furthermore, at 1200 B.C.E. we detect the first direct evidence for horse milk consumption, demonstrating the Bronze Age origins of equine dairying on the eastern steppe which occurred concomitantly with early evidence for horse bridling and riding in the region. The incorporation of horse milk and riding into early Mongolian subsistence strategies led to dramatic economic and demographic shifts that enabled the formation of the well-known steppe empires.
