SYSTEMS ANALYSIS OF THE LIVER TRANSCRIPTOME IN ADULT MALE ZEBRAFISH EXPOSED TO THE NON-IONIC SURFACTANT NONYLPHENOL (NP).

ABSTRACT: We report the effects of exposure to the endocrine disruptor nonylphenol (NP) on transcriptome modification in the livers of in vivo Zebrafish. Our data indicate changes in fatty acid metabolism and inflammation, pathways associated with the development of Non-Alcoholic Fatty Liver Disease (NAFLD).

Project description:Alterations in gene expression in FVB/NJ mouse liver following 4-nonylphenol (4-NP) treatment for 32 weeks. Mice were treated with 45 mg/kg/day 4-NP for 32 weeks, livers excised and differential gene expression determined. Samples 1 and 2 are controls and samples 3 and 4 are mice treated with 4-NP. Arrays were visualized from the K-screen, and analyzed and quantified with a phosphoimager (Molecular Imager FX, Bio-Rad, Hercules, CA) and the ResGen PathwaysTM Universal Microarray Analysis SoftwareTM (Research Genetics, Carlsbad, CA). Local background was subtracted automatically from the individual spot intensities, and spot intensities were normalized to the mean intensity of all spots on the array. This is called datapoint normalization on the ResGen Pathways Microarray Analysis SoftwareTM. Normalized gene intensities on the arrays probed with 4-NP treated mouse livers and untreated mouse liver cDNA were compared, and fold differences were calculated. Keywords: ordered

Transcriptomic Analysis of Nonylphenol Effect on Saccharomyces cerevisiae

Project description:Nonylphenol (NP), is a bioaccumulative environmental estrogen that is widely used as a nonionic surfactant. We have previously examined short-term effects of NP on yeast cells using microarray technology. In the present study, we investigated the long-term impacts of NP on Saccharomyces cerevisiae BY4742 cells by analyzing genome-wide transcriptional profiles using RNA-sequencing. We used 2 mg/L NP concentration exposure for 40 days. Gene expression analysis showed that a total of 948 genes were differentially expressed. Of these, 834 genes were downregulated, while 114 genes were significantly upregulated. GO enrichment analysis revealed that 369 GO terms were significantly affected by NP exposure. Further analysis showed that many of the differentially expressed genes were associated with oxidative phosphorylation, iron and copper acquisition, autophagy, pleiotropic drug resistance and cell cycle progression and related processes such as DNA and mismatch repair, chromosome segregation, spindle checkpoint activity, and kinetochore organization. Overall, these results provide considerable information and a comprehensive understanding of the long-term effects of NP at the gene expression level.

Microarray analysis among non-alcoholic fatty liver, non-alcoholic steatohepatitis, and type 2 diabetes mellitus rats

Project description:Non-alcoholic fatty liver (NAFL) has the potential to progress to non-alcoholic steatohepatitis (NASH) or to promote type 2 diabetes mellitus (T2DM). However, NASH and T2DM do not always develop coordinately. We established rat models of NAFL, NASH, and NAFL + T2DM to recapitulate different phenotypes associated with NAFLD and its progression. Microarrays were used to identify hepatic gene expression changes in each of these models. The goal is to identify a predictor of different NAFLD progressions. Non-alcoholic fatty liver disease (NAFLD) is recognized as a low-grade systemic inflammatory state with both hepatic and extra-hepatic manifestations. We aimed to identify common key regulators and adaptive pathways in different NAFLD phenotypes. NAFL, NASH and NAFL+T2DM rat models were used to represent simple fatty liver, fatty liver with severe hepatic manifestations, and fatty liver with severe metabolic manifestations, respectively. We applied microarray analysis to characterize the key regulators and adaptive pathways in different NAFLD phenotypes. There are 12 samples in our study which belonged to 4 groups, and each group contains 3 different samples.

We took advantage of human pluripotent stem cells (hIPSCs) to establish 3D cultures of hIPSCs-derived hepatocytes to study NAFLD pathogenesis in vitro. Cultures were challenged with free fatty acid to mimic NAFLD, and transcriptomic analyses were used to confirm the presence of a NAFLD signature.
