RNA-Seq expression profiling of mouse DEN-initiated hepatocytes with or without Tgfbr2 inactivation

ABSTRACT: Initiated hepatocytes (IHCs), isolated from DEN exposed Tgfbr2 flox/flox mouse, were infected with GFP or Cre-expressed adenovirus, followed by 1ng/ml TGFβ × 48 hours incubation. Then total RNAs were isolated and processed to routine sequencing on Illumina platform. RNA-Seq libraries were prepared from total RNA using polyA enrichment.

Project description:Transcriptional profiling of mouse prostate stromal cells knocked out for Tgfbr2 were compared to that of the Tgfbr2-flox control. Elevated chemokine expression in the prostate stroma of a castrate resistant mouse model, Tgfbr2-fspKO, prompted us to determine the role of bone marrow derived cell recruitment to the prostate during regrowth. Tgfbr2-flox prostate epithelia vs. Tgfbr2-fspKO prostae epithelia. Biological replicates: 3 Tgfbr2-flox mouse prostates pooled, 3 Tgfbr2-fspKO mouse prostate replicates.

Transcriptional profiling of mouse prostate tumors with conditional mutations in the Pten, Apc, or Tgfbr2 genes

Project description:We report the changes in gene expression in mouse prostate comparing normal wild type prostate to tumors generated by Cre mediated deletion of Pten or Apc, either with or without deletion of Tgfbr2. Pten single mutant tumors were isolated at either 8 weeks or 22 weeks of age, with high grade prostate intraepithelial neoplasia (HGPIN) as the main phenotype. Pten;Tgfbr2 double mutants were isolated at 8 weeks (primarily HGPIN), or at 11-14 weeks with extensive locally invasive cancer. Apc single mutants were isolated at 36 weeks old with adenosquamous HGPIN, or at 21-24 weeks of age for the Apc:Tgfbr2 double mutants, which had adenosquamous carcinoma. Ages for the two double mutant groups were based on initial signs of excess tumor burden. Wild type control prostates were isolated at around 20 weeks of age.

Murine livers post DEN treatment: c-Jun f/f vs. c-Jun Dli [43MM]

Project description:Liver carcinogenesis induced by DEN closely mimics the pathologies observed during human liver tumor initiation and progression. DEN treatment triggered c-Jun expression lasting up to at least 1 week in mouse livers as measured by Western blots and immunohistochemical staining. DEN-induced liver tumorigenesis was also dramatically reduced in Alfp-cre+/c-jun f/f mice, in which c-Jun is constitutively deleted in hepatocytes around birth. These data unambiguously demonstrate that c-Jun is specifically required during liver cancer initiation. We searched for cell death effectors downstream of c-Jun operating in cancer initiation. Expression profiling revealed that several cell death-associated genes were deregulated by comparing Alfp-cre+/c-junf/f and Alfp-cre-/c-jun f/f livers. Mouse livers were collected before and 48hours after diethylnitrosamine treament (100mg/kg body weight). Whole genome expression profiles were analyzed using total RNAs extracted from the whole liver samples.
