Project description:Heart failure (HF) is a global concern, marked by limited therapeutic options. While existing studies primarily focus on cardiomyocytes, there is a notable absence of drugs targeting noncardiomyocytes for HF. We focused on cardiac fibroblasts (CFs), utilising single-cell RNA-sequencing analysis. The analysis of murine hearts revealed one subcluster exclusive to the HF stage. The transcription factor c-Myc is specifically expressed in heart failure-specific fibroblasts (HF-Fibro). Cardiac fibroblast-specific deletion of c-Myc ameliorates pressure overload-induced cardiac dysfunction without affecting fibrosis. To elucidate the molecular function of c-Myc in HF-Fibro, transcriptome analysis by RNA-seq was conducted, in vivo.

Project description:Orangutans are an endangered species whose natural habitats are restricted to the Southeast Asian islands of Borneo and Sumatra. For potential species conservation and functional genomics studies, we derived induced pluripotent stem cells (iPSCs) from cryopreserved skin fibroblasts obtained from captive orangutans. We report the gene expression profiles of iPSCs and skin fibroblasts derived from orangtuans. The overall goal was to evaluate gene expression biomarkers of pluripotency in iPSCs and skin fibroblasts derived from PBD-ZSD patients and healthy controls. Dermal fibroblast cultures from 2 orangutans were reprogrammed into iPSCs by transfection with retroviruses designed to express the human OCT4, SOX2, KLF4 and c-MYC cDNA. Fibroblasts and iPSCs were cultured in 1:1 ratio of DMEM:F12 medium supplemented with 20% KOSR (knock-out serum replacement) at 37°C with 5% CO2 until confluence for RNA extraction. The overall goal was to evaluate gene expression biomarkers of pluripotency in iPSCs and original fibroblast cultures.

Project description:Oncoproteins of the MYC family drive the development of numerous human tumors. In unperturbed cells, MYC proteins bind to virtually all active promoters and control transcription by RNA Polymerase II (RNAPII). MYC proteins can also coordinate transcription with DNA replication and promote the repair of transcription-associated DNA damage, but how they exert these mechanistically diverse functions is unknown. Here we show that MYC dissociates from many of its binding sites in active promoters and forms multimeric, often sphere-like structures in response to perturbation of transcription elongation, mRNA splicing or inhibition of the proteasome. Multimerization is accompanied by a global change in the MYC interactome towards proteins involved in transcription termination and RNA processing. MYC multimers accumulate on chromatin immediately adjacent to stalled replication forks and surround FANCD2, ATR and BRCA1 proteins, which are located at stalled forks. MYC multimerization is triggered in a HUWE1- and ubiquitylation-dependent manner. At active promoters, MYC multimers block antisense transcription and stabilize FANCD2 association with chromatin. This limits DNA double-strand break formation during S phase, suggesting that multimerization of MYC enables tumor cells to proliferate under stressful conditions.

Project description:Cancer Associated Fibroblasts (CAF) are a dominant and critical cell type of the tumour microenvironment and can lead to breast cancer progression. TGF-β has been reported to influence fibroblast to myofibroblast activation, which is similar to cancer associated fibroblast phenotype. To understand the mechanism of TGF-β mediated CAF phenotype, we have performed a comparative proteomic analysis of conditioned media from CAF (isolated from breast cancer biopsy tissues) and normal mammary fibroblasts engineered to over-express TGF-β1. Liquid chromatography/ tandem mass spectrometry (LC ESI Q-TOF-MS/MS) assay of conditioned media and Venn analysis of the acquired data, revealed approximately 185 common proteins secreted by the three fibroblast types (CAF, normal mammary fibroblasts and TGF-β over expressing mammary fibroblasts). Among these, 12 proteins exclusively overlap between normal fibroblasts and CAF and 20 proteins exclusively overlap between CAF and TGF-β1 over-expressing fibroblasts. This analysis reveals interesting targets which may be important in activation phenotype of CAF and breast cancer progression.
