Project description:Branching morphogenesis is a basic way of the kinds of complex organs’ development including lungs, kidneys, thyroid glands and salivary glands.1 Submandibular gland (SMG) development is a classic model to reveal the mechanism of branching morphogenesis and it also provides novel approaches to tissue engineering for salivary glands regenerating or for creating artificial salivary glands.And for murine salivary glands’ development, two main components are formed, epithelial and mesenchyme. The elongation and branch of epithelial make up the main process of its development which supported by mesenchymal secretion.This processes are controlled by many factors like growth factors, hormones and miRNAs etc.567 All these factors play different roles in the branching morphogenesis of mouse salivary glands. Transforming growth factor β1 (TGF-β1) is a pivotal factor of them and it has a huge impact on SMG development through its action on the mesenchyme. We used microarrays to detail the global programme of gene expression during the branching morphogenesis and identified distinct classes of up/down-regulated genes during this process.

Project description:Development of branching organs involves a series of events regulated by proliferation, differentiation, and intricate cell-cell communication networks. Understanding the molecular drivers for these developmental stages is instrumental to device effective regenerative therapies. Here, we use single cell RNAseq of murine submandibular gland (SMG) from developmental stages corresponding to end bud initiation (E12), branching morphogenesis (E14), cytodifferentiation (E16) and postnatal development (P1, P30, and adult) to generate an atlas of SMG development. The generated resource highlights the heterogeneity in the salivary gland epithelium throughout development and allowed us to identify putative transcription factors involved in secretory acinar cell specification that occurs around E16, including Ybx1, Eno1, Atf3, and Atf4. Clustering and trajectory inference (pseudotime) analyses of this atlas illustrate and suggest that end bud and acinar populations represent the most differentiated state relative to other populations from the same developmental stage, which was associated with a global decrease in expression of numerous transcription factors. Lastly, we identified and characterized two subsets of intercalated duct cells, a Kit+ population defined by Gfra3, and a subset characterized by Gstt1 with sexually dimorphic Smgc co-expression in females and high Serpinb11 co-expression in males. Our analysis indicates that the Gstt1+ population originates from proacinar precursors suggesting plasticity in this population. This atlas can be combined with other available resources to learn about specific cell functions and to predict mechanisms involved in development of branching organs.

Project description:Salivary gland-specific binding assays reveal that CrebA, a bZIP transcription factor, directly binds the vast majority of genes encoding the secretory machinery, including proteins of the signal recognition particle and receptor, proteins involved in co-translational import of cargo into the ER, proteins involved in vesicular transport between the ER and Golgi, as well as the structural proteins and enzymes of these organelles. CrebA does not bind salivary gland-specific cargo genes. Instead, it binds and boosts expression of Sage, which encodes a bHLH transcription factor that upregulates cargo expression. CrebA also directly binds and upregulates Xbp1, which encodes a key factor in the unfolded protein response, and Tudor-SN, which encodes a protein that in other systems increases secretory cargo mRNA levels.

The long-term maintenance of diverse salivary gland cells in organoids remains problematic. Here, we established long-term murine salivary gland organoids from 3 major salivary glands, including parotid gland (PG), submandibular gland (SMG), and sublingual gland (SLG). Murine salivary gland organoids expressed gland-specific genes and proteins of acinar, myoepithelial, and duct cells. Organoids were maintained in growth media (named GEM) and further underwent differentiation in differentiation media (named DAM). Our study will provide an experimental platform for the exploration of mechanisms involvled in tissue regeneration, development, or several salivary gland diseases.

Project description:Chronic graft-versus-host disease (cGVHD) targets include the oral mucosa and salivary glands after allogeneic hematopoietic stem cell transplant (HSCT). Without incisional biopsy, no diagnostic test exists to confirm oral cGVHD. Consequently, therapy is often withheld until severe manifestations develop. This proteomic study examined saliva and human salivary gland for a biomarker profile at first onset of oral cGVHD prior to initiation of topical steroid therapy. Whole saliva collected at onset of biopsy-proven oral GVHD was assessed using liquid chromatography coupled tandem mass spectrometry with identification of 569 proteins, of which 77 significantly changed in abundance. ZG16B, a secretory lectin protein, was reduced 2-fold in oral cGVHD saliva (p < 0.05), and significantly decreased in salivary gland secretory cells affected by cGVHD. Single-cell RNAseq analysis of healthy MSG localized ZG16B expression to two discreet acinar cell populations. Reduced expression of ZG16B may indicate specific cGVHD activity, general salivary gland dysfunction.
