Characteristics and localization of prospective fibroblast subpopulations isolated from the mouse lung

ABSTRACT: By fluorescence-activated cell sorting, we classified lung fibroblasts highly expressing Pdgfra into three subpopulations, Sca-1highThy-1low (A-type), Sca-1highThy-1high (B-type), and Sca-1lowThy-1low (C-type). We then performed microarray analysis to identify specific genes for each subpopulation.

Metabolically distinct subpopulations of a secondary AML cell line are associated with differences in stemness and drug response

Project description:In recent years fatty acid metabolism has gained greater attention in haematological cancers, such as myeloid dysplastic syndrome (MDS) and secondary acute myeloid leukaemia (sAML), and has been implicated in increased proliferation and drug resistance. We have characterised two populations of FACS-sorted MDS/sAML cells - SKK-1LOW and SKK-1HIGH - that retain distinct fatty acid uptake phenotypes that are stable over time. Transcriptomic analysis of SKK-1LOW and SKK-1HIGH confirmed differentially expressed genes. Gene ontology analysis revealed gene expression differences principally in fatty acid transport genes and receptor tyrosine kinases, of which KIT was associated with low fatty acid uptake. SKK-1LOW resembled a leukaemic stem cell sub-population on the level of its immunophenotype and exhibited greater resistance to the hypomethylating agent, azacitadine. Overall, this study sheds new light on the relatively under-characterised SKK-1 cell line, which may be useful for the study of MDS-to-sAML transformation.

Constitutive activation of oncogenic PDGFR-mutants found in GIST patients induces receptor mislocalisation and alters PDGFR-A signalling characteristics

Project description:Gastrointestinal stomal tumors (GIST) are mainly characterised by the presence of activating mutations in either of the two receptor tyrosine kinases c-KIT and Platelet-derived growth factor receptor-a (PDGFRa). Most mechanistic studies dealing with GIST mutations have focused on c-KIT and far less is known about the signalling characteristics of the mutated PDGFRa proteins. Comparative studies of the signal transduction capacities of different mutant proteins cannot be performed in patient cells due to the patient specific background which may influence the signalling behaviour of the investigated mutants. In addition, the lack of a real wild-type signalling control (PDGFRa-WT) hampers comparisons of the signalling of wild-type and mutant proteins. Therefore, we stably and inducibly expressed different PDGFRa mutants as well as wild-type PDGFRa on an isogenic background in 293T cells in order to study the signalling capacities and corresponding transcriptional responses of the different proteins. We found that the constitutive signalling via the oncogenic PDGFRa mutants favours a mislocalisation of the receptors and that this modifies the signalling characteristics of the mutated receptors. We show that signalling via the oncogenic PDGFRa mutants is not solely characterised by a constitutive activation of the conventional PDGFRa signalling pathways.

Orchestrated intron retention regulates normal granulocyte differentiation [Affymetrix arrays]

Project description:Using mRNA-seq, we determined intron retaining genes that were differentially regulated in FACS purified cells at three progressive stages of mouse granulopoiesis; CD34+Kit+Gr-1low promyelocytes, CD34-Kit-Gr-1mid myelocytes and CD34-Kit-Gr-1high granulocytes. We found that IR affects 86 genes, including those specific to granulocyte (Lyz2 and MMP8) and nuclear architecture (Lmnb1 and Lbr). IR was associated with the decrease in protein levels measured by mass spectrometry (P=0.0015, binomial test). Inhibition of NMD in granulocytes resulted in marked accumulation of 39/86 intron retaining mRNAs (P<0.05, RUV procedure with Holm-Bonferroni correction), indicating that IR triggers NMD to downregulate mRNA and protein expression.

Single cell RNA sequencing of Hic1 and PDGFRa-labeled cells in the heart

Project description:The cardiac stroma contains multipotent mesenchymal progenitors. However, lineage relationships within cardiac stromal cells are poorly defined. Here, we identify heart-resident PDGFRa+ SCA-1+ cells as cardiac Fibro/Adipogenic Progenitors (cFAPs) and show that they respond to ischemic damage by generating SCA-1- fibrogenic cells. Pharmacological blockade of this differentiation step with an anti-fibrotic tyrosine kinase inhibitor decreases post-myocardial infarction (MI) remodeling and leads to improvement in heart function.
