Inhibition of SF3B1 by molecules targeting the spliceosome in Rh18 cells

ABSTRACT: In order to examine the effects of sudemycin D1 on the whole transcriptome, we exposed Rh18 cells to 1μM of drug for 8hr and after extraction, subjected RNA to RNAseq analysis. Control samples were treated with the vehicle (DMSO) alone and all analyses were conducted in triplicate.

Project description:HpeG2 cells were exposed in triplicate to three agrichemicals for 24hrs at 8 concentrations and a DMSO vehicle control (0.001, 0.003, 0.01, 0.03, 0.1, 0.3, 1, 3, and 10 μM plus DMSO vehicle controls). While a common set of DMSO controls was used, these CEL files were RMA normalized independently with each of the chemical treated groups. Project description:Gene expression changes in human OCI-AML3 cells treated with DMSO (vehicle control), the BET inhibitor CPI-203, the MDM2 inhibitor Nutlin-3a, or a combination of both drugs, were compared. Method: poly-A mRNA from the OCI-AML3 cell line treated for 24 hrs with either DMSO, CPI-203, Nutlin-3a or a combination of both drugs was extracted, in triplicate, then sequenced with an Illumina NextSeq500 sequencer.
