Characterization of miRNomes in two mouse Embryonic Stem Cell lines subjected to differentiation

ABSTRACT: We used high throughput miRNA sequencing to investigate differences between two mESC lines (D3 and B8 ESCs). Undifferentiated ESCs, cells differentiating in embryoid bodies (day 7), and also cells from embryoid bodies outgrowth (day 21) were analyzed.

Project description:Mouse Primordial Germ Cell Like Cells (mPGCLCs) are an in vitro model of the specification of mPGCs which happen around E6.25 in the developing mouse embryo. The aim of this study is the detection and profiling of mouse small RNAs via small RNA sequencing of E14-mESC-derived and Blimp1-GFP (BG5)-mESC-derived Day 6 mPGCLCs, each from diffent mouse strain backgrounds, alongside Day 6 non-PGCLC cells from E14, BG5 and Stella-GFP/Esg1-tdTomato (SGET) Embryoid Bodies. Project description:Mouse 129-B13 ESCs were genetically modified using CRISPR-Cas9 with two guides to remove exon 5 of Phf14, single-cell sorted and expanded to establish modified cell lines (2). Samples were differentiated from mouse ESCs (Day 0) to embryoid bodies (Day 4), neural progenitor cells (Day 8) and glutamatergic neurons (Day 12). Additionally, the parental 129-B13 ESC wild-type cells were split into two and differentiated separately as controls. NEBNext Poly(A) mRNA Magnetic Isolation Module was used to enrich mRNA. Nonsense-mediated decay was observed in Phf14 ex 5 KOs and no Phf14 protein expression was detected by western blot in these cells.
