Project description:Calcium is a universal second messenger molecule which plays a significant role in several biological processes. Presence of calcium sensors (calmodulins) and calcium-dependent protein kinases in Plasmodium species suggests an important role of calcium-dependent signaling pathways in the regulation of cellular processes in the malaria parasites. Evidence for the transcriptional response of Plasmodium falciparum asexual blood stages to the well-known calcium ionophore A23187 has been presented here. Comparative genomic hybridization (CGH) was carried out with total DNA isolated from the untreated parasites and the parasites treated with the calcium ionophore (A23187 5 μM concentration) at 1 hour, 4 hours and 6 hours post treatment. Genomic DNA from the isolates was extracted by phenol chloroform. 3 μg of the total DNA from each of the isolate was subjected to klenow(NEB) reaction as described in Bozdech Z, Llinas M, Pulliam BL, Wong ED, Zhu J, DeRisi JL: The transcriptome of the intraerythrocytic developmental cycle of Plasmodium falciparum. PLoS Biol 2003, 1(1):E5. Treated DNA labeled with Cy3 was then hybridized against untreated DNA labeled with Cy5.

Evidence for the transcriptional response of Plasmodium falciparum asexual blood stages to the well-known calcium ionophore A23187 has been presented here. P. falciparum 3D7 strain was cultured as described by Bozdech Z, Llinas M, Pulliam BL, Wong ED, Zhu J, DeRisi JL: The transcriptome of the intraerythrocytic developmental cycle of Plasmodium falciparum. PLoS Biol 2003, 1(1):E5. Calcium ionophore treatment was done as follows. Parasites at schizont stage were treated with 5 μM of calcium ionophore, A23187 for 30 minutes, 1 hour, 2 hours, 4 hours and 6 hours. Total RNA from each of the time points was isolated and aminoallyl-cDNA was synthesized using reverse transcriptase system (Fermentas). cDNA made from the treated parasites were labeled with Cy5 (GE-Amersham). A reference pool was made by mixing equal amount of cDNA from the parasites collected at 6 hours interval throughout the 48 hours life cycle and was labeled with Cy3 (GE-Amersham). The samples were then hybridized on a spotted cDNA chip platform comprising 10166 MOEs representing 5363 coding sequences as described in Hu G, Cabrera A, Kono M, Mok S, Chaal BK, Haase S, Engelberg K, Cheemadan S, Spielmann T, Preiser PR, Gilberger TW, Bozdech Z: Transcriptional profiling of growth perturbations of the human malaria parasite Plasmodium falciparum. Nat Biotechnol, 2009. 28(1): p. 91-8. The data was normalized and filtered with the condition, signal intensity>background intensity + 2 SD of background intensity) using NOMAD.

Project description:The purpose of the present study is to identify gene signatures with IgE/antigen and calcium ionophore A23187 on the activation of MMCs. The differentially expressed genes between the two types of samples were identified with the Microarray Analysis. The gene ontology functional and pathway enrichment analyses of differentially-expressed genes were performed using the database for annotation, visualization and integrated discovery software.

Project description:Ionophores are small molecules or peptides that transport metal ions across biological membranes. Their transport capabilities are typically characterized in vitro using vesicles and single ion species. It is difficult to infer from these data which effects ionophores have on living cells in a complex environment (e.g. culture medium), since net transport is influenced e.g. by ion composition, concentration gradients, pH gradient, and active transport. To gain insights into the antibacterial mechanism of the semi-synthetic polyether ionophore 4-Br-A23187, we investigated its effects on the gram-positive model organism Bacillus subtilis. Changes in intracellular ion concentrations were determined using ionomics and the physiological impact was assessed by proteomics. 4-Br-A23187 treatment resulted in an increase in intracellular copper levels, an effect that was dependent on the copper concentration of the medium. The antibiotic effect of 4-Br-A23187 is further aggravated by a decrease in intracellular manganese and magnesium. Effects of copper hyperaccumulation, mirrored by the proteomic response, included oxidative stress, disturbance of proteostasis, metal, and sulfur homeostasis. Especially solvent-exposed iron-sulfur clusters like that of aconitase of the citric acid cycle are readily destabilized by copper. Using calcein fluorescence quenching as a readout, we confirmed in vitro that 4-Br-A23187 acts as copper ionophore.

Project description:Purpose: To identify downstream genetics targets of the Dfi1 pathway during Dfi1 activating conditions. Methods: WT and dfi1 null cells were grown in minimal media, then treated with either 4uM of the calcium ionophore A23187 or an equal volume of 100% ethanol vehicle. After 30 min of treatment, cells were collected for RNA extraction and analysis. RNA was extracted using the Qiagen RNAeasy kit, then sent to the Tufts University Core Facility for library prep and sequencing. Results: Analysis of this data resulted in the identification of a number of possible targets of the Dfi1 pathway, as reported in the manuscript "Bypass of Dfi1 regulation of Candida albicans invasive filamentation by iron limitation".

Project description:The Scott syndrome is a rare bleeding disorder associated with a mutation in the gene encoding anoctamin-6 (TMEM16F). After stimulation of Ca2+-mobilizing agonists, syndromatic platelets show a reduced phosphatidylserine exposure and do not form membrane blebs. Given the central role of anoctamin-6 in the platelet procoagulant response, we used quantitative proteomics to understand the underlying molecular mechanisms and the complex phenotypic changes in Scott platelets compared to control platelets. Therefore, we applied an iTRAQ-based multi-pronged strategy to quantify changes in (i) the global proteome, (ii) the phosphoproteome and (iii) proteolytic events between resting and stimulated Scott and control platelets. Our data indicate a limited number of proteins with decreased (70) or increased (64) expression in Scott platelets, among those we observed the absence of anoctamin-6 and the strong up-regulation of aquaporin-1. Furthermore, the quantification of 1,566 phosphopeptides revealed major differences between Scott and control platelets after stimulation with thrombin/convulxin or ionomycin. Finally, we quantified 1,596 N-terminal peptides in activated Scott and control platelets, 180 of which we identified as calpain-regulated, whereas a distinct set of 23 neo-N-termini was caspase-regulated. In Scott platelets, calpain-induced cleavage of cytoskeleton-linked and signaling proteins was down-regulated, in accordance with an increased phosphorylation state. Thus, multi-pronged proteomic profiling of Scott platelets provides detailed insight into their protection against detrimental Ca2+-dependent changes that are normally associated with phosphatidylserine exposure.

Project description:MRP14 is tissue-specific expressed only in granulocytes, monocytes and in inflammation-activated keratinocytes. Its existence in the early phase of inflammation may be a hint towards a participation in immune response. Intra- and extracellular functions have been described yet. An antibacterial effect is caused by chelation of zinc, which then cannot be used for bacterial growth. The experiments were performed to shed light on the function of the SA100 protein MRP14. Experiment Overall Design: MRP14(SA100A9)-positive and -negative mouse bone marrow granulocytes are measured unstimulated as control (2 repeats each at time point 0h, 5 repeats each at time point 4h) and then stimulated for 4h with LTB-4 (1 repeat each), A23187 (a calcium ionophore, 4 repeats each), LPS (part of gram negative bacteria, 3 repeats each) and PMA (1 repeat each); MRP14(SA100A9)-positive and -negative mouse spleen tissue (1 repeat each) for comparison; another data set contains all experiments of this data set with Mu11KsubA instead of Mu11KsubB arrays
