Gene expression analysis of human fetal pancreatic niche cell lines: Wk17.5h and Wk20.1

ABSTRACT: We derived cell lines from the human pancreatic niche from different sites during different temporal stages of fetal development when beta cells are differentiating and maturing. This screen uncovered differences between the human pancreatic niche at fetal week 17.5 head of pancreas(Wk17.5h) and 20.1 doudeum (Wk 20.1) and control cell line human dermal fibroblast(HDF), illustrating a temporally changing, developmentally significant microenvironment.

MicroRNA profiling of human dermal fibroblasts - miR-17 and miR-19b to investigate down-regulatation in human aging

Project description:Normal human fibroblasts were isolated from skin biopsies of two different healthy causcasian patients, a 41-years old female (HDF-1) and a 16-years old male (HDF-5). HDF-5 and HDF-1 cells were cultivated in DMEM/HAMs F-12 medium (Biochrom KG, Berlin, Germany) supplemented with 10% fetal calf serum (FCS) 4 mM L-glutamine (Sigma) and Primocin antibiotics (100 µg/mL) grown at 37 °C in ambient atmosphere containing 5% CO2. Cells were grown until confluence was reached and routinely passaged using 1:4 or 1:3 split ratios. Upon decrease in growth rate and entry of irreversible growth arrest split ratio was changed to 1:2 until cells stopped dividing and fresh medium was added at least every 7 days. HDF-1 cells reached senescence after 54 population doublings, while HDF-5 cells had stopped growing after 72.7 population doublings (see Fig. S2 for growth curves and staining of senescent associated beta-galactosidase).<br>Senescent HDF RNA samples were compared to young HDF RNA samples in terms of microRNA expression using Exiqon LNA microRNA microarrays.

SOX9+/PTF1A+ Cells Define the Tip Progenitor Cells of the Human Fetal Pancreas of the Second Trimester

Project description:Significant progress has been made in recent years in characterizing human multipotent progenitor cells (hMPCs) of the early pancreas; however, the identity and persistence of these cells during the second trimester, after the initiation of branching morphogenesis, remain elusive. Additionally, studies on hMPCs have been hindered by few isolation methods that allow for the recovery of live cells. Here, we investigated the tip progenitor domain in the branched epithelium of human fetal pancreas between 13.5 and 17.5 gestational weeks by immunohistological staining. We also used a novel RNA‐based technology to isolate live cells followed by gene expression analyses. We identified cells co‐expressing SOX9 and PTF1A, two transcription factors known to be important for pancreatic MPCs, within the tips of the epithelium and observed a decrease in their proportions over time. Pancreatic SOX9+/PTF1A+ cells were enriched for MPC markers, including MYC and GATA6. These cells were proliferative and appeared active in branching morphogenesis and matrix remodeling, as evidenced by gene set enrichment analysis. We identified a hub of genes pertaining to the expanding tip progenitor niche, such as FOXF1, GLI3, TBX3, FGFR1, TGFBR2, ITGAV, ITGA2, and ITGB3. YAP1 of the Hippo pathway emerged as a highly enriched component within the SOX9+/PTF1A+ cells. Single‐cell RNA‐sequencing further corroborated the findings by identifying a cluster of SOX9+/PTF1A+ cells with multipotent characteristics. Based on these results, we propose that the SOX9+/PTF1A+ cells in the human pancreas are uncommitted MPC‐like cells that reside at the tips of the expanding pancreatic epithelium, directing self‐renewal and inducing pancreatic organogenesis.

Proteomic analysis of laser-microdissected murine fetal glomeruli

Project description:Alteration of fetal kidney morphogenesis in diabetic pregnancies is poorly described, especially changes in the extracellular matrix (ECM) and glomerular basement membrane (GBM) during glomerulogenesis. Addressing the ECM proteome, or matrisome, in mouse fetal glomeruli in a healthy and diabetic environment will improve understanding about the association between ECM and GBM changes in the glomerulus as potential reprogramming mechanisms for glomerular dysfunction in infants of mothers with diabetes. This project aimed to define the matrisome and BM composition in the maturing murine fetal glomeruli collected from normal and diabetic mothers. For this, we used laser microdissection microscopy to isolate maturing glomeruli from cryosections, and analyse by label-free tandem mass spectrometry to test the hypothesis that maternal diabetes influences ECM and GBM composition, assembly and function.

Integrated small RNA, mRNA and protein omics reveal a miRNA network orchestrating metabolic maturation of the developing human heart

Project description:As the fetal heart develops, cardiomyocyte proliferation potential decreases while fatty acid oxidative capacity increases, a highly regulated transition known as cardiac maturation. Small noncoding RNAs, such as microRNAs (miRNAs), contribute to the establishment and control of tissue-specific transcriptional programs. However, small RNA expression dynamics and genome wide miRNA regulatory networks controlling maturation of the human fetal heart remain poorly understood. Transcriptome profiling of small RNAs revealed the temporal expression patterns of miRNA, piRNA, circRNA, snoRNA, snRNA and tRNA in the developing human heart between 8 and 19 weeks of gestation. Our analysis revealed that miRNAs were the most dynamically expressed small RNA species throughout mid-gestation. Cross-referencing differentially expressed miRNAs and mRNAs predicted 6,200 mRNA targets, 2134 of which were upregulated and 4066 downregulated as gestation progresses. Moreover, we found that downregulated targets of upregulated miRNAs predominantly control cell cycle progression, while upregulated targets of downregulated miRNAs are linked to energy sensing and oxidative metabolism. Furthermore, integration of miRNA and mRNA profiles with proteomes and reporter metabolites revealed that proteins encoded in mRNA targets, and their associated metabolites, mediate fatty acid oxidation and are enriched as the heart develops.This study revealed the small RNAome of the maturing human fetal heart. Furthermore, our findings suggest that coordinated activation and repression of miRNA expression throughout mid-gestation is essential to establish a dynamic miRNA-mRNA-protein network that decreases cardiomyocyte proliferation potential while increasing the oxidative capacity of the maturing human fetal heart.

Proteomic analysis underlying the impact of early life nutrition on the molecular control of testicular development in the young bull calf.

Project description:It has been established that enhanced early life nutrition progresses sexual development in the bull calf through neuroendocrine signalling via the hypothalamic-pituitary-testicular axis. However, the underlying molecular mechanisms regulating this process have not been fully elucidated. This study measured the impact of contrasting feeding regimes in the first 12 wk of life, known to impact age at puberty, on the proteomic landscape of the testes of bull calves. Holstein bull calves with a mean (±SD) bodyweight and age of 48.8 (± 5.3) kg and 17.5 (± 2.8) days, were designated to high (HI; n=10) or moderate (MOD; n=10) dietary groups, with diets designed to provoke growth rates of 1.0 and 0.5 kg/day, respectively. At 12 wk of age, all calves were euthanized, and testes parenchyma harvested. HI calves were heavier at slaughter (112.4 v 88.7 (2.98) kg, P<0.001), and had a greater average daily gain (ADG) of (0.88 v 0.58 kg, P<0.001). The turquoise network from the protein analyses contained the protein CDH13 which is involved in testes development. Gene ontology analysis of the turquoise network revealed enrichment of genes with functions related to cholesterol biosynthesis, IGF-1 signalling, insulin receptor/secretion signalling, androgen signalling and Sertoli cell junction signalling.
