MiRNA Expression Profiles of Premalignant and Malignant Arsenic-Induced Skin Lesions

ABSTRACT: Total RNA was purified from keratinocytes isolated from FFPE arsenic-induced skin lesion samples collected from individuals exposed to high concentrations of arsenic exceeding 50 ppb in drinking water in Murshidibad district of West Bengal, India.

Chronic Arsenic Exposurre Impairs Adaptive Thermogenesis in Male C57BL/6J Mice

Project description:Purpose: To determine the effects of sodium arsenite in male mice on adaptive thermogenesis. Methods: Male C57BL/6J mice were exposed to sodium arsenite in drinking water at 300 parts per billion (ppb) for 9 weeks Findings: Arsenic-treated mice experienced significantly decreased metabolic heat production when acclimated to chronic cold tolerance testing, as evidenced by indirect calorimetry, despite no change in physical activity. Arsenic exposure increased total fat mass, and unilocular lipid droplet size in both subcutaneous inguinal white adipose tissue (iWAT) and brown adipose tissue (BAT). Conclusion: Chronic arsenic exposure impacts the mitochondria of thermogenic tissues involved in energy expenditure and glucose regulation, providing novel mechanistic evidence for arsenic’s role in metabolic pathologies.

Whole genome transcriptional analysis in rice-stem during dough stage induced by arsenic contaminated soil and irrigation water

Project description:RNA from stem containing first node and internodes during dough stage was considered for the above purpose. Satabdi, (popularly known as Minikit) most popular cultivar of West Bengal and Palman, another high yielding variety were considered for microarray analysis for comparison to each other. Two rice genotypes were grown in identical conditions (same field with contaminated ground water) to obtain the most meaningful conclusion. Dough stage was considered as variation in arsenic accumulation among the genotypes starts during dough stage to maturity. Plant tissues were collected from stem for transcriptomic analysis as expression profiling of such tissue in response to arsenic would be most useful as arsenic transport/loading in grain was assumed to be controlled by arsenic translocation behavior at inter vascular level or from xylem to phloem. Satabdi accumulates more than twice of arsenic in brown rice (0.346 mg/kg) than that of Palman (0.156 mg/kg) whereas accumulation in straw (2.07 mg/kg) was approximately less than half of the Palman (4.491 mg/kg)

Project description:Adult male mice (F0) were exposed to 250 ppb inorganic arsenic (iAs) in drinking water before mating with unexposed female mice to generate male F1 offspring (iAsF1-M). Unexposed male mice were bred simultaneously to generate male controls (conF1-M). Both iAsF1-M and conF1-M mice drank normal water without iAs. Adult iAsF1-M and conF1-M mice were harvested to collect liver samples to do RNA-seq.

Project description:Russell’s viper (Daboia russelii) (RV), a category I medically important snake as well as a member of the “Big Four”, is responsible for a heavy toll of snake bite mortality and morbidity in Indian sub-continent. Epidemiological studies suggest highest incidence of RV envenomation in eastern India (EI). In this study the RV venom proteomes from Burdwan and Nadia, the two districts of West Bengal, eastern India was deciphered for the first time using tandem mass spectrometry analysis.

Defining effects of in utero arsenic exposure on immune cells in lungs of adult mice exposed to Influenza A with single cell RNA sequencing

Project description:Our recent studies have shown that adult mice exposed to inorganic arsenic exclusively in utero (100 ppb via maternal drinking water) exhibit greater lung damage and inflammation following infection with IAV (Influenza A virus, H1N1) than control mice not exposed in utero. We employed an unbiased single cell RNA sequencing approach to conduct a high resolution molecular analysis of recruited immune cells and their transcriptional activities in the lungs of arsenic-exposed and control mice at day 3 after IAV infection. Transcriptional profiling of single immune cells from 6 mice (3 As, 3 Con, all exposed to IAV) revealed evidence of an enhanced inflammatory response mediated by dysregulation of innate immune functions of monocyte derived macrophages, neutrophils and alveolar macrophages.

Transcription profiling of mouse lung from animals exposed to arsenic

Project description:Exposure to high levels of arsenic in drinking water is associated with several types of cancers including lung, bladder and skin, as well as vascular disease and diabetes. Drinking water standards are based primarily on epidemiology and extrapolation from higher dose experiments, rather than measurements of phenotypic changes associated with chronic exposure to levels of arsenic similar to the current standard of 10ppb, and little is known about the difference between arsenic in food as opposed to arsenic in water. Measurement of phenotypic changes at low doses may be confounded by the effect of laboratory diet, in part because of trace amounts of arsenic in standard laboratory chows, but also because of broad metabolic changes in response to the chow itself. Finally, this series contrasts 8hr, 1mg/kg injected arsenic with the various chronic exposures, and also contrasts the acute effects of arsenic, dexamethasone or their combination. Male C57BL/6 mice were fed on two commercially available laboratory diets (LRD-5001 and AIN-76A) were chronically exposed, through drinking water or food, to environmentally relevant concentrations of sodium arsenite, or acutely exposed to dexamethasone. Experiment Overall Design: Male C57BL/6 mice, fed on two commercially available laboratory diets (LRD-5001 and AIN-76A), were chronically exposed through drinking water or food, to environmentally relevant concentrations of sodium arsenite. Project description:In utero exposure to arsenic via drinking water increases the risk of lower respiratory tract infections during infancy and mortality from bronchiectasis in adulthood. Pregnant mice were exposed to arsenic, and gene expression patterns were profiled in peripheral lung tissue obtained from the offspring at 2 weeks of age. Pregnant Mice (BALB/c, C57BL/6, C3H/HeARC) mice were exposed to arsenic (or control) via drinking water from day 8 of gestation until the birth of their offspring. After giving birth, mothers were given control drinking water. At 2 weeks post-natal age, total RNA was extracted from peripheral lung tissue of the offspring and analysed on Affymetrix microarrays.
