Project description:In mammals, temporally coordinated daily rhythms of behaviour and physiology are generated by a multi-oscillatory circadian system, entrained through cyclic environmental cues (e.g. light). Presence of niche-dependent physiological time cues have been proposed to allow local tissues flexibility of adopting a different phase relationship if circumstances require. Up till now, such tissue-unique stimuli have remained elusive. Here we show that cycles of mechanical loading and osmotic stimuli within physiological range drive rhythmic expression of clock genes and reset clock phase and amplitude in cartilage and intervertebral disc tissues.

Effect of cyclic tensile stretch on meniscus cells in the presence and absence of IL-1

Project description:Meniscus injuries are highly prevalent and are linked to the development of post-traumatic osteoarthritis (PTOA). The inflammatory cytokine IL-1 is elevated in synovial fluid following knee injuries, causes degradation of meniscus tissue, and inhibits meniscus repair. Cyclic tensile stretch has been shown to modulate the expression of pro-inflammatory cytokines and degradative enzymes induced by IL-1; however, there remains a dearth of knowledge on global effects of loading on meniscus cell phenotype and transcriptomic profiles. In this study, we performed mRNA-Seq on meniscus cells isolated from inner and outer zone regions of porcine menisci subjected to cyclic tensile strain in the presence and absence of IL-1 to identify cellular responses to mechanical load, identify differences in response to load based on zonal characteristics, and identify IL-1 induced inflammatory responses modulated by load.

Project description:Understanding how cells respond to and cope with high levels of mechanical stress is important to gain a better understanding of mechano-biology, both in health and disease. The experiment was designed to assess the total mRNA levels in mechanically stimulated cells and was used, in conjunction with mass spectrometry data, to gain an insight into the systemic response to high-intensity mechanical strain. Here, total RNA was extracted from human mesenchymal stem cells (n=6) following 1 hour of cyclic tensile strain (2.6 – 6.2% strain, 5.0 Hz) using a Flexcell Tension Plus device. RNA from unstrained cells was used as control. The protein coding RNAs were then sequenced with Illumina HiSeq technology.

Cyclic tensile strain enhances the expression of Indian hedgehog and progression of ossification of the human cervical posterior longitudinal ligament

Project description:The initiation and/or progression of ossification of the posterior longitudinal ligament (OPLL) is associated with cyclic tensile strain, but the pathomechanism of OPLL remains unclear. Indian hedgehog (Ihh) and its related signaling are key factors in normal enchondral ossification. However, the relation of OPLL to Ihh is unclear. The purpose of this study is to investigate the contribution of mechanical strain to OPLL and the relation of Ihh to OPLL.

RNA-seq of Runx2 cKO primary chondrocytes.

Project description:Purpose: To demonstrate Runx2's association with organizing the extracellular matrix in primary chondrocytes. Method: RNA samples were collected from primary chondrocytes of 5-day-old Col2a1-CreERT2;Runx2fl/fl and Runx2fl/fl mice, cultured with or without IL-1beta. Results: Thirty-three genes cultured without IL-1beta and 45 genes with IL-1beta were up- or down-regulated by more than 2-fold in the Runx2 knockout in primary chondrocytes. Conclusions: Genes including collagen fibers were down-regulated in Runx2 cKO primary chondrocytes.

Method: Fragmented DNA samples were collected from primary chondrocytes of 5-day-old Runx2-Biotin-FLAG-tag mice, cultured with or without IL-1beta. Results: More than 20,000 and 10,000 peaks were gained from chondrocytes without and with IL-1beta, resepectively. Conclusions: Runx2 are associated cellular process and extracellular matrics transcription in primary chondrocytes.

Charaterising the surfaceome of primary equine chondrocytes

Project description:The aim of this study was to characterize the surfaceome of primary equine chondrocytes isolated from healthy joints following exposure to the pro-inflammatory cytokines interleukin-1-beta (IL-1β) and tumour necrosis factor-alpha (TNF-α). We employed combined methodology that we recently developed for investigating the surfaceome in stem cells. Membrane proteins were isolated using an aminooxy-biotinylation technique and analysed by mass spectrometry using high throughput shotgun proteomics. Amongst the 431 unique cell surface proteins identified, a high percentage of low-abundance proteins, such as ion channels, receptors and transporter molecules were detected. A high number of proteins exhibited different levels of expression following chondrocyte stimulation with pro-inflammatory cytokines. LPR-1, thrombospondin, VDAC1-2 and annexin A1 were chosen for further analysis and validation by western blotting as proteins of special interest. Our results provide, for the first time, a repository for proteomic data on differentially expressed low-abundance membrane proteins on the surface of chondrocytes in response to pro-inflammatory stimuli.
