Transcriptional Analysis of Foxp3+ Regulatory T Cells in Experimental Acute Lung Injury Resolution

ABSTRACT: These studies profiled the expression of mRNA in lung and spleen regulatory T cells in Foxp3EGFP mice during lipopolysaccharide-induced lung injury. Baseline, uninjured Lung Treg and Resolving Lung Tregs and Resolving Splenic Tregs were sorted (7 days after intratracheal instillation of LPS for Resolving conditions) and mRNAs differentially expressed (DE) between Control Lung Tregs, Resolving Lung Tregs and Resolving Splenic Tregs samples were identified using microarrays.

Project description:We isolated total lung RNA from spontaneously hypertensive male rats 2â40 h after exposure to reference EHC-93. Three animals were exposed to EHC-93 in saline(10 mg/kg body weight) or saline at each time via intratracheal instillation or no instillation at all. RNA from different times was pooled for array hybridization as indicated.

Project description:To investigate the role of H2S in Chronic obstructive pulmonary disease (COPD), a rat model of COPD was established by cigarette smoking (CS) and intratracheal instillation of lipopolysaccharide (LPS). Rats were randomly divided into 4 groups: Control, CS+LPS, CS+LPS+NaHS and CS+LPS+propargylglycine (PPG). Transcription profiling of lung tissue comparing Control with CS+LPS, CS+LPS with CS+LPS+NaHS, and CS+LPS with CS+LPS+PPG. The goal was to determine the impact of H2S on gene expression profiling in rat model of COPD.

Mice were intra-nasally treated with lipopolysaccharide (LPS, 20 μg/mouse) to induce pulmonary inflammation and subsequently exposed to B[a]P (0.5 mg/mouse) by intratracheal instillation We used RNA microarrays to detail the global gene expression change after each treatment. Meanwhile, compared the up- and down-regulated gene between the LPS&BaP and BaP in order to identify the role of inflammation on BaP induced carcinogenesis
