NFAT3-regulated genes in human aortic smooth muscle cells (ASMC)

ABSTRACT: NFAT family transcription factors play crucial roles in immunological and other biological activities. Since we found that NFAT3 was highly expressed in ASMC, the target genes of NFAT3 in ASMC were investigated.

Gene sequencing analysis of mouse airway smooth muscle cells in co-culture environment with mouse airway epithelial cells and mouse smooth muscle cells in non-co-culture environment

Project description:Airway epithelial cells can promote the proliferation of airway smooth muscle cells in an undisturbed environment. We isolated airway epithelial cells and airway smooth muscle cells in mice and conducted co-culture experiments. The results showed that the proliferative phenotype of airway smooth muscle cells was significantly changed by co-culture of epithelial cells after five day. Therefore, we conducted gene sequencing for ASMC in the co-culture group and ASMC in the non-co-culture group after 5 days of culture. The results show that Wnt pathway related genes are activated in ASMC, and the activation of this pathway may be the main reason why AEC stimulates the proliferation of ASMC

Expression data from asthamtic and healthy airway smooth muscle cells

Project description:Persistent severe asthma is associated with hyper-contractile airways and structural changes in the airway wall, including an increased airway smooth muscle (ASM) mass. This study used gene expression profiles from asthmatic and healthy airway smooth muscle cells grown in culture to identify novel receptors and pathways that potentially contributed to asthma pathogenesis. We used microarrays to compare the gene expression between asthmatic and healthy airway smooth muscle cells to understand the underlying pathway contributing the differences in cellular phenotypes Asthmatic airway smooth muscle cells (ASMC) are intrinsically different and have a differential transcriptional response to pro-fibrotic, pro-proliferation and pro-inflammatory stimuli than ASMC from healthy patients. We sought to identify genes that are differentially expressed between asthmatic and healthy ASMC under various stimulations which mimic the asthmatic airways. To this end, we obtained human ASMC from bronchial biopsies and explanted lungs from doctor diagnosed asthmatic patients (n=3) and healthy controls (n=3). The ASMC were then grown in culture and treated with pro-fibrotic (Transforming growth factor beta (TGFβ)), pro-proliferation (Fetal Bovine Serum (FBS)) and pro-inflammatory stimuli (Interleukin-1 beta (IL-1β)) for 8 hours. Gene expression was then evaluated using Affymetrix Human Gene 1.0ST arrays.

RNA-seq analysis of pulmonary arterial smooth muscle cells (PASMCs) and bronchial smooth muscle cells (BSMCs) after pulmonary arterial hypertension (PAH) treatment

Project description:Previous studies to determine transcriptional changes in PASMCs during PAH were affected by the inevitable contamination with BSMCs and venous SMCs. To specific target ASMCs, we generated an ASMC-effector mouse line by combining Cspg4/Acta2 intersectional genetics, enabling specific targeting of vascular SMCs, including PASMCs. To target non-vascular SMCs, including BSMCs, we generated the NVSMC-effector mouse line using Chrm2/Acta2 intersectional genetics. The development of ASMC-effector mice, excluding venous and BSMCs, enabled us to avoid such problems and determine PASMC-specific reactions after induction of PAH.

VSMC were treated with AngII with or without CsA ( as an inhibitor of the CN/NFAT pathway). 4 independent VSMC cultures (4 replicates) from aortas of C57BL/6 mice were used for each condition (1uM AngII or 1uM AngII+200ng/ml CsA).

Project description:To identify distal chromatin contacts between promoters and their putative regulatory elements in human airway smooth muscle cells (ASMC), we performed Hi-C with a capture step to enrich the library for contacts involving promoters. Promoter capture Hi-C was performed according to PMID: 29988018 The cells were cultured and maintained in 10% FBS in PBS for 24 hours with no additional treatment given. probes-MboI_fragments-hg19.baitmap.bz2 can be used in CHiCAGO to call interactions. Interactions provided correspond to hg19 coordinates.
