Role of IL-17RA in the development of parasite-specific cytotoxic responses during Trypanosoma cruzi infection

Project description:IL-17RA knockout mice presented increased tissue parasitism during T. cruzi infection that correlated with a reduced frequency of parasite-specific CD8+ T cells. Parasite-specific CD8+ T cells from KO mice showed similar proliferation rate but increased mortality and an exhausted phenotype when compared to WT counterparts. We hypothesize that during T. cruzi infection, IL-17RA signaling in CD8+ T cells is required for the regulation of a transcriptional program that prevents accelerated mortality and exhaustion and sustain the development of robust cytotoxic response required for efficient parasite control.

Transcriptome profile of Trypanosoma cruzi-infected cells

Project description:As Trypanosoma cruzi, the etiological agent of Chagas disease, multiplies in the cytoplasm of nucleated host cells, infection with this parasite is highly likely to affect host cells. We performed an exhaustive transcriptome analysis of T. cruzi-infected HeLa cells using an oligonucleotide microarray containing probes for greater than 47,000 human gene transcripts. In comparison with uninfected cells, those infected with T. cruzi showed greater than threefold up-regulation of 41 genes and greater than threefold down-regulation of 23 genes. Real-time reverse transcriptase-polymerase chain reaction (RT-PCR) of selected, differentially expressed genes confirmed the microarray data. Many of these up- and down-regulated genes were related to cellular proliferation, including seven up-regulated genes encoding proliferation inhibitors and three down-regulated genes encoding proliferation promoters, strongly suggesting that T. cruzi infection inhibits host cell proliferation, which may allow more time for T. cruzi to replicate and produce its intracellular nests. These findings provide new insight into the molecular mechanisms by which intracellular T. cruzi infection influences the host cell, leading to pathogenicity. Keywords: infection response

Crucial role of intrinsic IL-18R/MyD88 signaling and IL-12 for Th1 and cytolytic CD4+ T cell responses during infection with the human parasite Trypanosoma cruzi.

Project description:We compared gene expression between WT CD4+ (CD45.1) and Myd88-/- CD4+ (CD45.2) T cells from the spleen of infected mix bone marrow chimeras at day 14 of T. cruzi infection. The hypothesis studied in this experiment was that MyD88-/- CD4+ cells have suppressed Th1 differentiation potencial when compared to WT CD4+ T cells. The results indicate that CD4+ T cell-intrinsic MyD88 signaling is necessary for sustaining a more robust Th1 differentiation program and increased expansion among WT CD4+ T cells, resultant of relative higher proliferation and lower apoptosis Total RNA obtained from isolated CD4+ T cells from the spleen of infected mix bone marrow chimeras at day 14 of T. cruzi infection.

Transcriptome analysis of the Trypanosoma cruzi life-cycle

Project description:To generate a high quality, annotated gene expression database of T. cruzi trypomastigotes(TRP), amastigotes (AMA), epimastigotes (EPI), and metacyclic trypomastigotes (MET). The global assessment of transcript abundances in each life-cycle stage of T. cruzi will identify stage-regulated genes and provide an essential element of an integrated database of T. cruzi genomic, proteomic, and transcriptomic data. RNAs from 3 biological replicates from a single time point in each stage will be subjected to DNA microarrays containing probes for the complete, annotated T. cruzi genome, as it is currently known. The arrays for this project will be provided by the Pathogen Functional Genomics Resource Center (PFGRC) at The Institute for Genomic Research (TIGR) sponsored by the National Institute of Allergy and Infectious Diseases (NIAID), and will contain long oligonucleotides complementary to every annotated gene in the newly sequenced T. cruzi genome. Pancreatic epithelial IL-17/IL-17RA signaling drives B7-H4 to promote tumorigenesis

Project description:IL-17 is required for the initiation and progression of pancreatic cancer, particularly in the context of inflammation, as previously shown by genetic and pharmacological approaches. The cellular compartment and downstream molecular mediators of IL-17-mediated pancreatic tumorigenesis have not been fully identified. We interrogated the cellular compartment required by generating transgenic animals with Interleukin 17 receptor A (IL-17RA) genetically deleted from the pancreatic epithelial compartment vs. the hematopoietic compartment via generation of IL-17RA-deficient (IL17-RA-/-) bone marrow chimeras, in the context of embryonically activated or inducible Kras. Deletion of IL-17RA from the pancreatic epithelial compartment, but not from hematopoietic, resulted in delayed premalignant lesions initiation and progression and increased CD8+ cytotoxic T cells infiltration to the tumor microenvironment. Absence of IL-17RA in the pancreatic compartment affected transcriptional profiles of epithelial cells, modulating stemness and immunological pathways. Interestingly, B7-H4, a known inhibitor of T cell activation encoded by the gene Vtcn1, was the most upregulated checkpoint molecule via IL17 early during pancreatic tumorigenesis, and its genetic deletion delayed pancreatic premalignant lesions development and reduced immunosuppression. We reveal pancreatic epithelial IL-17RA requirement for pancreatic tumorigenesis by reprogramming the immune pancreatic landscape which is partially orchestrated by regulation of B7-H4.
