Differential gene expression in TREM1 -/- vs +/+ myeloid cells 48h after MCAo-RP

ABSTRACT: Stroke is a multiphasic progress characterized by neuron damage due to hypoxia followed by secondary damage from the subsequent inflammatory immune response. Infiltrating myeloid cells induce cerebral damage through pro-inflammatory cytokines, chemokines, proteases and generation of reactive oxygen species (ROS). Triggering receptor expressed on myeloid cells 1 (TREM1) is exclusively expressed on myeloid cells and acts as an amplifier of pro-inflammatory innate immune responses. Using microarray analysis, we aim to identify the expression differences and functional change of these myeloid cells after TREM1 knockout in mice post stroke.

Identification of TREM1+CD163+ myeloid cells as a deleterious immune subset in HCC [Spatial Transcriptomics]

Project description:Hepatocellular carcinoma (HCC) is an inflammation-associated cancer arising from viral and non-viral etiologies. Expansion of suppressive myeloid cells is a hallmark of chronic inflammation and cancer, but their heterogeneity in HCC is not fully resolved and might underlie immunotherapy resistance in the steatohepatitis setting. Here, we present a high resolution atlas of hepatic innate immune cells from patients with HCC that unravels a steatohepatitis contexture characterized by influx of inflammatory and immunosuppressive myeloid cells. A discrete myeloid cell population identified by selective expression of TREM1 and CD163 expands in steatohepatitis-HCC. We refer to this population as TREM1+ regulatory myeloid cells (Mreg), as it potently suppresses T cell effector functions, highly expresses TGFB1 and IL13RA1 and localizes to HCC fibrotic lesions.

Project description:To gain a global understanding of the impact of TREM1 silencing, we analyzed the CD45+ tumor infiltrating cells (TICs) of B16F10 tumor-bearing Trem1+/+ and Trem1-/- mice. Utilizing the 10x Genomics Chromium Platform, we analyzed approximately 5390 cells per sample with a coverage rate of 15493 genes per cell.

Transcription profiling of human monocytes after treatment vehicle, isotype control antibody, TREM1 antibody, LPS, isotype control antibody plus LPS and TREM1 antibody plus LP to invesigate innate immune responses to TREM-1 activation

Project description:TREM-1 is an orphan immunoreceptor expressed on monocytes, macrophages, and neutrophils. TREM-1 associates with and signals via the adapter protein DAP12/TYROBP, which contains an immunoreceptor tyrosine-based activation motif (ITAM). TREM-1 activation by receptor cross-linking is pro-inflammatory, and can amplify cellular responses to Toll-like receptor (TLR) ligands such as bacterial lipopolysaccharide (LPS). To investigate the cellular consequences of TREM-1 activation, we have characterized global gene expression changes in human monocytes in response to TREM-1 cross-linking in comparison to and combined with LPS. Both TREM-1 activation and LPS up-regulate chemokines, cytokines, matrix metalloproteases, and PTGS/COX2, consistent with a core inflammatory response. However, other immunomodulatory factors are selectively induced, including SPP1 and CSF1 (i.e., M-CSF) by TREM-1 activation and IL-23 and CSF3 (i.e., G-CSF) by LPS. Additionally, cross-talk between TREM-1 activation and LPS occurs on multiple levels. While synergy in GM-CSF protein production is reflected in commensurate mRNA abundance, comparable synergy in IL-1b protein production is not. TREM-1 activation also attenuates the induction of some LPS target genes, including those that encode IL-12 cytokine family subunits. Whereas positive TREM-1 outputs are abolished by the PI3K inhibitor wortmannin, this attenuation is largely PI3K-independent. These experiments provide a detailed analysis of the cellular consequences of TREM-1 activation, and highlight some of the complexity in signal integration between ITAM- and TLR-mediated signaling. Project description:Analysis of Kupffer cells at gene expression level. The hypothesis tested in the present study was that the expression of TREM-1 on Kupffer cells isolated from wild type mice subjected to single dose of carbon tetracloride and sacrificed 12 hours after injection influences the expression of multiple genes implicating in several signaling pathways. Results provide important information of the response of Trem1-sufficient and Trem1-deficient Kupffer cells to carbon tetracloride, such as inflammatory responsive genes, up-or-down regulated inflammation, acute liver injury and liver fibrogenesis.
