The immune defense of shrimp gills through Marsupenaeus japonicus gill C-type lectin (MjGCTL)

ABSTRACT: To present, the only known inv*olvement of the gills in the immune response of shrimp is solely assisting the hemocytes in filtering out the harmful factors. This global expression of novel genes revealed several immune-related genes specifically expressed in high amounts only in gills. This data provide new insights on the immune defense of shrimp.

Project description:There is continuous interest in the anti-viral immune response of shrimp to prevent bacterial and viral infection. In this study, global expression analysis through microarray was conducted to gain insights about the mechanisms involved in the protection of non-specific double stranded RNA –injected shrimps against infectious diseases. Hemocytes were collected from different shrimp groups injected with PBS (Phosphate buffered saline), siGFP and dsGFP at 12 and 24 hour timepoints. Genes were clustered based on expression and treatment to highlight differences in expression related to the type of injection.

Shrimp transcriptome analysis after exposure to recombinant Vibrio parahaemolyticus PirA and PirB toxins

Project description:Vibrio parahaemolyticus is a Gram-negative bacterium commonly found in marine and estuarine environments. Acute hepatopancreatic necrosis disease (AHPND) caused by this bacterium is an ongoing problem among shrimp farming industries. V. parahaemolyticus proteins PirA and PirB have been determined to be major virulence factors that induce AHPND. In this study, Pacific white shrimp (Litopenaeus vannamei) were challenged with recombinant PirA and PirB by a reverse gavage method and then at 30 m, 1, 2, 4, and 6 h time points, the hepatopancreas of five individual shrimp were removed and placed into RNA later. We conducted RNA sequencing of the hepatopancreas samples from a no PirA/B control (n = 5) and PirA/B-treated shrimp at the different time intervals (n=5). We evaluated the different gene expression patterns between the time groups to the control with a focus on identifying differences in innate immune function.

Project description:The effect of dietary immunostimulation in the portals of entry, intestine and gills, of rainbow trout (Oncorhynchus mykiss), was investigated using a salmonid-specific microarray platform enriched with immune-related genes. IS-diet feeding significantly changed transcriptomic expression profiles: larger reduction rather than induction was observed, with significant changes in genes and functional GO categories related to remodeling processes and antigen presentation. The results revealed that one of the main effects of IS-diets in trout is the increase of genes involved in antigen recognition in epithelial cells of gills. The gills transcriptome analysis of yellow catfish (Tachysurus fulvidraco♀ × P. vachellii♂) under transport stress

Project description:Transport stress is a big threat to most teleost fish during production practice, causing mass loss to the aquaculture industry. Fish gills are mucosa-associated lymphoid tissue that directly contacted with water, which is the good choice to study the mechanism of transport stress. Gills transcriptome results revealed 1551 differentially expressed genes (744 up-regulated and 807 down- regulated) between the control and 16h transportation groups. The top 10 enriched KEGG pathways are all immune-related. After qRT-PCR validation, we found that the toll-like receptors and nod-like receptors signaling pathways mediate the gill’s immune response to transport stress: tlr5, tnfɑ, hsp90ɑ, il-1ß and map2k4 were significantly up-regulated and arrdc2 was significantly down-regulated under transport stress. These results suggested that the transport stress altered innate immunity responses and induced metabolic changes in gills of hybrid yellow catfish. In conclusion, the results provide new perspectives on immune response in yellow catfish against transport stress and provide the possibility of mining resistance genes for future optimize transportation plan.
