The miRNA expression signatures in young and aging C57BL/6 mouse kidney

ABSTRACT: To identify novel aging-related miRNAs, we initially established a physiological aging mouse model (20-month old male C57BL/6 mouse), compared with 2-month old male C57BL/6 mouse. Then, the Agilent miRNA microarray was performed to profile miRNA expression levels in kidney from 20-month old male C57BL/6 mouse (designated as Aging) and 2-month old male C57BL/6 mouse (designated as Young).

Bcl11a is essential for B-1a cell maintenance during aging (RNA-Seq)

Project description:B-1a cells are important immune cells, serving as the first line of defense against pathogens. B-1a cell undergo a series of alterations during aging and decrease the protection to our body. However, the characteristics of B-1a cells during the aging process are not fully understood. In this study, we discrible transcriptional and epigenetic profiles of B-1a from 3-month-old and 24-month-old mice across both genders. Interestingly，We found that the expression of the transcription factor Bcl11a was positively correlated with the number of B-1a cells in aged male and aged female mice.To further characterize senescent B-1a cells and the mechanisms of Bcl11a regulates B-1a. ATAC-seq and RNA-seq were performed on the B-1a isolated from young male mice (3-month-old),old male mice (24-month-old mice), young female mice (3-month-old),old female mice (24-month-old mice) of C57BL/6,and Bcl11a delated young male mice. CUT&tag was performed on the B-1a isolated from young male mice. Collectively, we provide a comprehensive resource to decode the aging process of B-1a, shedding light on how Bcl11a regulate B-1a cells maintenance.

Project description:We performed an RNA Sequencing experiment on dorsal hippocampal tissue from six groups of animals: Aging (18-20-month-old) HDAC3flox/flox homecage (H3F-HC); Aging (18-20-month-old) HDAC3flox/flox 60min post training (H3F-BV); Aging (18-20-month-old) wildtype homecage (OWT-HC); Aging (18-20-month-old) wildtype 60min post training (OWT-BV); Young (2-4-month-old) wildtype homecage (YWT-HC); Young (2-4-month-old) wildtype 60min post training (YWT-BV). Homecage animals were sacrificed directly from the animal's cage. Behavior animals were sacrificed sixty minutes following a 10min Object Location Memory training session.

RNA-seq was used to analyze hepatic gene expression from mice on 1-month chow diet, 1-month HFCF diet, 3-month chow diet, and 3-month HFCF.
