Aurora kinase A is upregulated in cutaneous T-cell lymphoma and represents a potential therapeutic target

Project description:Cutaneous T-cell lymphomas form a heterogeneous group of non-Hodgkin lymphomas characterized by only poor prognosis in advanced stage. Despite significant progress made in the identification of novel genes and pathways involved in the pathogenesis of cutaneous lymphoma, the therapeutic value of these findings has still to be proven. Here, we demonstrate by gene expression arrays that aurora kinase A is one of highly overexpressed genes of the serine/threonine kinase in CTCL. The finding was confirmed by qualitative RT-PCR, Western blotting and immunohistochemistry in CTCL cell lines and primary patient samples. Moreover, treatment with a specific aurora kinase A inhibitor blocks cell proliferation by inducing cell cycle arrest in G2 phase as well as apoptosis in CTCL cell lines. These new data provide a promising rationale for using aurora kinase A inhibition as a therapeutic modality of CTCL. 14 lesional skin biopsies of different MF patients (patch=3, plaque=6, tumor=4) and 9 healthy controls. Ablation of casein kinase 1 in keratinocytes induces p53-dependent hyperpigmentation of the skin

Project description:To characterize the role of CK1 (encoded by Csnk1a1) in skin physiology, we crossed mice with floxed Csnk1a1 with mice expressing K14CreERT2 to generate mice in which tamoxifen induces the deletion of Csnk1a1 exclusively in keratinocytes (SKO). In addition, we established K14CreERT2 CK1/p53 double-knockout (DKO) to analyze the effect of coablation of both genes. 4-hydroxy-Tamoxifen was applied for 14 days to induce the deletion of CK1 and/or p53 in the epidermis of the mice. In addition, wild type mice where exposed to UVB irradiation to compare the effect of CK1a ablation with the effects of normal UV exposure. In comparison, we analyzed also wild type mice as reference. RNA was obtained from cells derived from mouse ear dorsal-epidermis and tail skin. Subsequently, we performed RNA sequencing and transcriptome analysis.

Transcriptional pathway signatures for the Aurora inhibitor Danusertib

Project description:Treatment with Aurora inhibitors has been shown to induce diverse biological responses in different tumor cell lines, in part depending on their p53 status. To characterize at the transcriptional level the effects of Danusertib we analyzed by microarray different tumor cell lines, with WT or mutant p53 status, that showed differential cell cycle response upon drug treatment. We analyzed the effects of Danusertib treatment in different tumor cell lines derived from ovary (A2780, p53WT), breast (MCF-7, p53WT and MDA-MB-468, p53 mut) and colon carcinoma (HCT116, p53 WT and Colo205, p53 mut). Cell line were treated (TRT) or left unreated (CTRL) for 24 hrs with 1 uM Danusertib.
