Project description:In order to characterize iHep cells more precisely, we conducted global gene expression analyses using microarrays to compare among the gene expression profiles of MEFs, iHep cells and adult mouse hepatocytes. Three MEF samples that were individually prepared from 13.5 days post coitum (dpc) embryos, three types of iHep cells and hepatocytes obtained from the adult mouse liver were used.

Project description:We found that hepatocytes isolated from adult mouse livers can dedifferentiate into progenitor-like cells and exhibit a hybrid epithelial/mesenchymal phenotype in monolayer culture. dediHeps can re-differentiate into mature hepatocytes by forming aggregates and fetal intestinal progenitor cells (FIPCs) by forming organoids in three-dimensional culture. We conducted RNA-seq analyses to investigate the characteristincs of hepatocytes, dediHeps, dediHep aggregates, dediHep derived spherical organoids, dediHep derived budding organoids, FIPC derived spherical organoids, intestinal stem cell (ISC) derived budding organoids, and CLiP cells (Katsuda, 2016).

Then we mapped 2 histone modification marks (H3K9ac and H3K27me3) and 2 states of Pol2 (with phosphorylated serine 5 and phosphorylated both of serine 5 and serine 2) during the conversion event. Besides we performed FAIRE-Seq to explore the chromatin accessbility in the iHep cells.

Three-dimensional (3D) cell culture models are recently expected to bridge the gap between 2D and in vivo models. We have developed novel 3D culture method that improves the growth of spheroid-forming cancer cells under anchorage-independent condition by leveraging a feature of FP001. Gene microarrays were used to observe the global gene expression in A549 cells cultured with normal adhesion plate (2D, control) or with low adhesion plate (+FP001) and identified distinct classes of up or down-regulated genes.
