Diurnal gene expression in rosette leaves of the phosphoglucomutase mutant (Col-0)

Project description:How do transcript levels of leaf-expressed genes change in a normal day-night cycle of the phosphoglucomutase (pgm) mutant? The interest is in genes that are regulated by the circadian clock and the diurnal component (i.e. light, metabolite changes). Plants were grown on soil in a 12/12 h light/dark rhythm at 20°C day and night. 5 weeks after germination the rosettes of the non-flowering plants were harvested, 15 plants per sample. Photooxidative stress and light acclimation in the PSII antenna-deficient ch1 mutant

Project description:cea10-02_light - photooxidative stress - To characterize the metabolic pathways implicated in oxidative stress responses and in acclimation mechanisms in the ch1 mutant - The extracts are carried out starting from sheets of Arabidopis thaliana having pushed on compost in controlled conditions (light: 250 µmol.m-2.s-1, temperature: 22°C day 18°C night, moisture: 55%, photoperiod: 8:00 jour/16h night) during 4 and 8 weeks for respectively the Col0 genotype and the mutant ch1. The acclimatization of the plants is done during 48 H under an average luminous intensity (450 µmol.m-2.s-1, photoperiod 8:00), whereas the stress requires a strong luminous intensity (900 µmol.m-2.s-1, photoperiod 8:00) and low temperature (10°C day, 14°C night) also during 48 hours.

Proteasome and deubiquitinase activity were inhibited at 10 am (2 h of light) by floating leaf discs on ½ Hoagland supplemented with 50 µM MG132 and 20 µM PR-619 for 5 h.

Salicylic acid-induced gene expression in wild-type Col-0 and mutant upl1, upl5, and npr1-1 Arabidopsis thaliana plants

Project description:The plant immune hormone salicylic acid (SA) modulates transcriptional reprogramming via controlling the master transcriptional coactivator, NPR1. Here we examined the role of HECT-type ubiquitin ligases, UPL1 and UPL5, in SA/NPR1-dependent transcriptional control. We showed that UPL1 and UPL5 are essential regulators of SA-responsive genes, and regulate SA-induced transcriptional reprogramming in a NPR1-dependent manner. Four-week old Arabidopsis thaliana plants of wild-type Col-0, mutant upl1, mutant upl5, and mutant npr1-1 genotypes were germinated on soil in 100% relative humidity. Plants were continuously grown in an environmental chamber with 16/8 hour day/night light regime (120 mol m-2 s-1 light intensity), 21/18 degrees celcius day/night cycle and 65% relative humidity. 4-week-old plants were sprayed with water or 0.5 mM SA until all leaves were thoroughly covered with fine droplets, samples were collected after 24 hours treatment. In total two independent biological repeats were collected.

The translatome of both genotypes after 2 hours of light (10 am) was determined by feeding of the trackable Met-analogue azidohomoalanine for 4 hours to leaf disks floated on ½ Hoagland medium containing 50 µM azidohomoalanine in the light.

Heat-adaptive proteome in filling wheat grain

Project description:Wheat (T. aestivum L. Chinese Spring) plants were cultured in 25-cm buckets at a density of 16 plants per bucket in a greenhouse at a temperature of 15–30 °C. Plants were watered once a week, and artificial light was applied when there was fewer than 12 h of natural daylight. The flowering date of each spike was recorded when the first flower appeared on the spike. Comparable wheat plants were selected depending on the flowering time and growth status and were transferred into growth chambers 12 DAF for a three-day adaptation period to the growth chamber environment (24/17 °C, 14/10 h day/night cycle). Wheat plants were illuminated with a photosynthetic photon flux density of 360 µmol m˗2 s˗1 at canopy height, and relative humidity ranged from 40% to 70%. Half of the wheat plants (about 50 plants) were transferred to heated conditions (37/17 °C, 14/10 h day/night cycle, 40% to 70% humidity), while the others were maintained under the standard conditions described above as a control. After three day/night cycles, the heated samples were collected after the plants had been exposed to heat for 4 h, and the control samples from the unheated chambers were collected at the same time for and proteomic analyses. A minimum of 20 spikes were collected from each condition, immediately immersed in liquid nitrogen and stored at ˗80 °C until further analysis.
