For that, we planned the study of the response of the transcriptome in the presence of exogenic L-carnitine. Seedlings WT are cultivated on medium MS, 6 independent times. At 144h, the seedlings are collected and pools are formed with 3 samples. We proceed in the same way for the seedlings which are cultivated on medium MS containing L-carnitine 5mM. Genome wide analysis of glucose brassinosteroid interaction in Arabidopsis

Project description:Arabidopsis thaliana seeds after imbibition were inoculated in ½ MS medium supplemented with 0.8% agar and 1% sucrose. Once the plant material was uniformly germinated, the experimental conditions were applied. 5d old light-grown uniformly germinated seedlings were washed seven times with sterile water with last wash given by ½ MS liquid medium without sucrose to remove residual exogenous sugar and the plant material was kept in ½ MS liquid without sucrose in the dark for all subsequent steps. Cultures were shaken at 140 rpm at 22oC for 24 h and then 3 h treatment was given with liquid ½ MS without glucose and liquid ½ MS supplemented with BR (0.1 ?M EBR), glucose (3%), glucose (3%) + BR (0.1 ?M EBR). Seedlings were harvested after 3h and preceded for RNA isolation and Microarray analysis.

Project description:mRNA sequencing was used to identify genes differentially expressed in Delta1-pyrroline-5-carboxylate synthase1-4 (p5cs1-4) mutants deficient in low water potential-induced proline accumulation and the Abscisic Acid (ABA) deficient mutant aba2-1 compared to Col-0 wild type. Three treatments were analyzed for each genotype: unstressed control, a 96 hour low water potential (-1.2 MPa) treatment and a stress-release treatment where seedlings were removed from the stress treatment and transferred back to control media for 10 hours. Approximately 100 genes were found to be significantly up-regulated in p5cs1-4 compared to wild type in both the control and stress treatments with an equal number down regulated. There was also a substantial transcription response to the stress release treatment which varied between wild type, p5cs1-4 and aba2-1. Wild type (Col-0), p5cs1-4 and aba2-1 Arabidopsis thaliana seedlings were germinated on agar plates and after seven days of growth, seedlings were transferred either to fresh plates of control media or to low water potential (-1.2 MPa) stress plates. Samples were collected 96 h after transfer (Control and stress treatments). At 96 h, additional seedlings were transferred from the stress treatment back to control agar plates and samples collected 10 h after this stress release. Three independent biological experiments and mRNA extractions were performed for each genotype/treatment (27 mRNA extractions. After quality check and quantitation, the mRNA samples were pooled to produce a set of 9 samples for sequencing.

Global gene expression analysis of Arabidopsis Col-0 under normal and low oxygen conditions

Project description:Global gene expression was compared between root RNA samples from three-week-old Arabidopsis Col-0 plants subjected to 0.1% oxygen (balance nitrogen) or ambient atmospheric conditions. Three-week-old Arabidopsis seedlings (4-6 leaves) of ecotype Col-0 were used. Two biological replicates were used for each treatment, and for each biological replicate, five Arabidopsis seedlings grown on the same petri dish were bulked. RNA was extracted from root tissue excised post-treatment. Treatment consisted of moving plants grown on solid MS media containing 3% sucrose to liquid MS media containing 3% sucrose for 24 hours, after which plates were placed in anaerobic cannisters covered in aluminum foil. Cannisters containing plants in the 0.1% oxygen treatment group were flushed with a 0.1% oxygen gas mixture and then sealed, while cannisters in the control group were left unsealed. Treatment lasted 5 hours before plant tissue was harvested and RNA extracted.
