Project description:In our previous work we substantiated the ability of the Fgd5-mCherry mice to tag hematopoietic stem cells (HSCs) during immune response (Bujanover et al., 2018). While most of the research is focused on viral-bacterial models mediated by a Th1 response less has been investigated on Th2 mediated hypersensitivity. Our current experimental model of allergic peritonitis seeks to understand the effect of a Th2 mediated immune response utilizing our transgenic Fgd5-mCherry mice. Our results showed no significant change in quantity of HSCs and their immediate progenitors in the bone-marrow post treatment. Furthermore, transplant experiments showed no significant change in the long term multipotent ability of HSCs vs. control. Examining the cell cycle state of HSCs, quiescence is an essential property for stemness, we find no evidence of proliferation. We continued to sort and bulk RNAseq HSCs after allergic peritonitis showing relatively small changes in expression profile. Taken together, this data suggests no prominent response of HSCs to our Th2-mediated hypersensitivity model.

Project description:Hematopoietic stem cells (HSCs) maintain the entire blood system throughout the life and are utilized for a therapeutic component of blood diseases. The zebrafish is an elegant genetic model for the study of hematopoiesis due to its many unique advantages. We have developed the method to isolate zebrafish HSCs by a combination of two HSC-related transgenic lines, gata2a:GFP and runx1:mCherry. In this study, we performed RNA-seq analysis in three distinct hematopoietic cell populations in the adult kidney, gata2a+ runx1+ cells (HSCs), gata2a− runx1+ cells (erythroid- and/or myeloid-primed progenitors), and gata2a+ runx1− cells (lymphoid-primed progenitors).

Project description:We compared transcriptomes of CD34(high)EPCR-positive with CD34(high)EPCR-negative cells after 10-days culture in PCL-PVAc-PEG based 3a medium. It was because our culture condition had not previously reported and so we demonstrated that phenotypic HSCs were concentrated in EPCR-positive fraction as conventional UM171-based culture system. As a result, CD34(high)EPCR-positive cells highly expressed HSC markers (HLF, AVP, PRDM16 and FGD5) compared with CD34(high)EPCR-negative cells.

Project description:Hematopoietic stem cells (HSCs) cultured outside the body are the fundamental component of a wide range of cellular and gene therapies. Recent efforts have achieved more than 200-fold expansion of functional HSCs, but their molecular characterization has not been possible due to the substantial majority of cells in these cultures being non-HSCs and the fact that single cell-initiated cultures display substantial clone-to-clone variability. Using the Fgd5 reporter mouse in combination with EPCR, we have identified an in vitro reporter strategy capable of exclusively identifying HSC-containing clones and prospectively separating HSCs from non-HSCs. Linking single clone functional transplantation data with single clone gene expression profiling, we also reveal the molecular profile of expanded HSCs which is more similar to actively cycling fetal liver HSCs than quiescent adult bone marrow HSCs from which they were derived. A gene expression signature derived from the molecular profiling identifies core molecular components of HSCs, including Prdm16, Fstl1 and Palld, that can further predict the functional outcome in transplantation assays. Taken together, this system represents a new tool to be used for a wide-range of functional screening and molecular experiments previously not possible to carry out due to limited HSC numbers

Project description:Adult and fetal hematopoietic stem cells (HSCs) display a glycolytic phenotype, which is required for maintenance of stemness; however, whether mitochondrial respiration is required to maintain HSC function is not known. Here we report that loss of the mitochondrial complex III subunit Rieske iron sulfur protein (RISP) in fetal mouse HSCs allows them to proliferate but impairs their differentiation, resulting in anemia and prenatal death. RISP null fetal HSCs displayed impaired respiration resulting in a decreased NAD+/NADH ratio. RISP null fetal HSCs and progenitors exhibited an increase in both DNA and histone methylation concomitant with increases in 2-hydroxyglutarate (2-HG), a metabolite known to inhibit DNA and histone demethylases. RISP inactivation in adult HSCs also impaired respiration resulting in loss of quiescence resulting in severe pancytopenia and lethality. Thus, respiration is dispensable for adult or fetal HSC proliferation, but essential for fetal HSC differentiation and maintenance of adult HSC quiescence.

Project description:This dataset is part of a study that investigated how the hematopoietic system coordinates the rapid and efficient regeneration of the megakaryocytic lineage during stress scenarios. We found that the phenotypic hematopoietic stem cell (HSC) compartment contains stem-like megakaryocyte-committed progenitors (SL-MkPs), a cell population that shares many features with multipotent HSCs and serves as a lineage-restricted emergency pool for inflammatory insults. This dataset contains single-cell RNA sequencing data of 30 hematopoietic stem and progenitor cells which, in the context of our study, confirmed that MK-specfic transcripts are of highly variable expression in HSCs. The dataset further showed that variations in MK transcript expression in HSCs is not correlated with global transcriptomic rearrangements. Murine bone marrow cells were sorted by Lin-cKit+CD150+CD48- (referred to as cd150+ in the following) and Lin-cKit+CD150- (referred to as cd150- in the following). Project description:Here we aim to dissect the complex nature of the acute systemic response of HSCs to lipopolysaccharide (LPS), mimicking a bacterial infection. Acute LPS challenge transiently induced proliferation of quiescent HSCs in vivo, without yet impairing HSC function. We hypothesized that the effect of LPS on HSCs is indirectly mediated via proinflammatory cytokines. Our results showed that Acute LPS exposure depends in part on IFN signaling to activate HSCs.

Project description:Hematopoietic stem cells (HSCs) are generated via a natural transdifferentiation process known as endothelial-to-hematopoietic cell transition (EHT). Due to small numbers of embryonal arterial cells undergoing EHT and the paucity of markers to enrich for hemogenic endothelial cells, the genetic program driving HSC emergence is largely unknown. Here, we use a highly sensitive RNAseq method to examine the whole transcriptome of small numbers of enriched aortic HSCs (CD31+cKit+Ly6aGFP+), hemogenic endothelial cells (CD31+cKit-Ly6aGFP+) and endothelial cells (CD31+cKit-Ly6aGFP-). Comparison of mRNA profiles of endothelial cells, hemogenic endothelial cells, and hematopoietic stem cells generated by deep-sequencing of sorted populations from pool of embryos, in triplicate.
