Project description:T2 progenies of two transgenic lines overexpressing ERF transcription factor WIN1 were grown on soil in parallel under identical conditions. mRNA was extracted from pooled leaves from multiple plants of each line for the microarray experiement. Plants derived from two lines were used in this experiment: line 13, a "medium" overexpressor, and line 22, a strong overexpressor. Control mRNA was obtained from pooled control plants transformed using an empty binary vector. All plants were first selected on medium containing kanamycin before being transferred to soil.

Arabidopsis has three MIXTA-like proteins NOECK(NOK)/MYB106, MYB16 and MYB17 and nok mutant exhibits over-branched trichomes in rosette leaves. On the other hand, Arabidopsis AP2/ERF transcription factor WAX INDUCER1/SHINE1 is well known to induce cuticle development. Here, we report the transcriptional cascade of MYBs-WIN/SHNs coordinately regulates the epidermal cuticle development. The constitutive expression of MYB106 chimeric repressor, in which strong repression domain was fused with MYB106 (35S:MYB106SRDX), induced cuticle deficiency characterized by organ adhesion, reduction of epicuticular wax crystals and staining by toluidine blue in broad region of aerial parts in addition to the over-branched trichomes. Most these phenotypes were partially reproduced in T-DNA knockout plants and also similarly induced by the chimeric repressors of WIN1/SHINE1(SHN1), SHN2 and SHN3, which are AP2/ERF transcription factors previously shown as positive regulators of cutin. Microarray experiments revealed that MYB106SRDX and WIN1SRDX plants have similar transcriptomes, in which the expression of wax and cutin biosynthetic genes was suppressed. Conversely, VP16-fused MYB106 which has high activation ability induced ectopic production of cutin nanoridges in the transgenic plants and enhanced the reporter driven by promoters of WIN1 and wax-related genes in transient expression assay. These results suggest that MIXTA-like proteins regulate cutin biosynthesis partially via WIN1 and directly regulate wax accumulation as well as their known roles in epidermal cell differentiation.

Project description:Potato cultivar Russet Norkotah was grown in a growth chamber at 16 hour light/8 hour dark at 22°C fertilized with Osmocote. Plants were from tissue culture, transplanted to soil and used approximately 4 weeks after transplanting. RNA was extracted from leaves using Trizol. Plants were sprayed with one of four treatments, either 1mM salicylic acid, 100 µM methyl jasmonate, 1 mg/ml arachidonic acid or 100 µM methyl jasmonate plus 1 mM salicylic acid together. Samples were harvested at 3, 24 and 72 hours after spraying. Each salicylic acid or arachidonic acid treatment has 3 biological replicates done in parallel, using leaves pooled from 2 plants. The control for SA or AA treated plants were water sprayed plants, from which leaves of 3 different plants were pooled at each timepoint, done side-by-side with the treated samples. Methyl jasmonate or MJ+SA treatments had 2 biological replicates each, using leaves pooled from 2 plants. A separate set of water sprayed plants (from the SA and AA control treatments) were used as controls, and pooled leaves from 3 plants were used at each time-point, and done in parallel with the treated samples. Project description:Although bodyguard (bdg), lacerata (lcr) and fiddlehead (fdh) mutations affect three unrelated genes, they trigger similar effects, i.e. ectopic organ fusion, increase of cuticle permeability. After performing cutin and wax analyses on these Arabidopsis thaliana mutants, which did not coincide with the putative enzyme functions, we hypothesised that these mutations trigger a complex response which may be visible at the transcriptional level. This analysis revealed that major changes in gene regulation are taking place in bodyguard (bdg), lacerata (lcr) and fiddlehead (fdh) mutants, massively affecting genes associated with lipid, cell wall and intracellular homeostasis but also with pathogen resistance. Experiment Overall Design: Plants carrying a stable allele of the bodyguard (bdg), lacerata (lcr) and fiddlehead (fdh) mutations, respectively, were grown on soil in parallel to wild type Columbia plants and under controlled growth conditions. Trays were periodically shifted to avoid any position effect and, to increase representativeness of the samples, each sample contains leaf tissue from 15 plants.

Project description:The 5th and 6th leaf blades of the rice Os-LBD37 overexpressor line RK16331-13 and the empty vector control line FOX3 were examined. LBD37 belongs to the plant- specific LOB- (Lateral Organ Boundary) domain family proteins first characterized in Arabidopsis. Results point towards an involvement of the rice LBD37 (OsLBD37) ortholog of Arabidopsis in nitrogen metabolism- and senescence- related processes. Hygromycin- resistant rice Os-LBD37 overexpressor and empty vector control plants were grown in hydroponic culture system. The sampling date of the leaf blades of RK16331-13 LBD37 overexpressor plants and the empty vector control was determined according to the emergence of the 8th leaf blade. Three independent hybridizations were performed for line RK16331-13 and the empty vector control line.
