Project description:Drug resistance and distant metastases are leading causes of mortality in colorectal cancer (CRC), yet the molecular mechanisms linking these processes remain elusive. In this study, we demonstrate that acquired resistance to oxaliplatin, a first-line chemotherapeutic in CRC, enhances metastatic potential through transcriptional reprogramming. Using a clinically relevant dosing regimen, we generated oxaliplatin-resistant CRC cells that displayed increased metastatic potential. Integrated transcriptomic and phenotypic analyses revealed that dysregulated cholesterol biogenesis amplifies TGF-b signaling, which in turn drives expression of SERPINE1, that serves as a key effector of both oxaliplatin resistance and metastasis. Furthermore, we uncovered a SERPINE1-associated nine-gene expression signature, RESIST-M, that robustly predicts overall and relapse-free survival across distinct patient cohorts. Notably, RESIST-M stratifies a high-risk subtype of CMS4/iCMS3-fibrotic patients that display the poorest prognosis, underscoring its clinical relevance. Targeting of SERPINE1 or cholesterol biosynthesis re-sensitized resistant, pro-metastatic cells to oxaliplatin in mouse xenograft models. Altogether, this study uncovers a mechanistic link between metabolic rewiring and transcriptional plasticity underlying therapy-induced metastasis in primary CRC. Additionally, it also reveals actionable vulnerabilities that offer both prognostic value and therapeutic potential.

Project description:In the present study, natural antisense transcripts (NATs), transcribed from reverse strand deoxynucleic acids (DNAs) of genes, into exosomes released from colorectal cancer SW480 cells were searched using an sense/antisense-custom microarray. SW480 cells were cultured with serum-free RPMI1640 for 48 h. Then, culture media were collected, centrifugated, and filtrated using 0.22-um filters. Exosomes from culture media were collected by ultracentrifugation.

Synergistic enhancement of anticancer effects by periodontal ligament stem cell exosomes and oxaliplatin via apoptosis induction and protein modulation

Project description:Periodontal ligament stem cell-derived exosomes (PDLSC-exosomes) exhibit promising therapeutic potential in cancer treatment. Here, we characterize PDLSC-exosomes and investigate their effects on colorectal (HT-29, HCT-116) and oral (SCC-25) cancer cell lines. Nanoparticle tracking analysis and transmission electron microscopy revealed exosome diameters ranging from 80-220 nm and typical cup-shaped morphology. Flow cytometry confirmed surface markers CD9, CD63, and CD81. PKH67-labeling demonstrated efficient exosome internalization by all three cell lines. PDLSC-exosomes significantly inhibited cancer cell growth and colony formation in a dose-dependent manner. Synergistic effects were observed when combined with oxaliplatin, enhancing growth inhibition, apoptosis induction, and motility reduction. Apoptotic assays revealed increased Bax and p53 gene expression and altered protein profiles, particularly those involved in apoptosis and immune signaling pathways. Proteomic analysis identified differentially expressed proteins linked to translational processes and vesicle fusion. This study highlights the synergistic effects of PDLSC-exosomes and oxaliplatin, providing novel insights into their potential as an adjunct therapy to enhance cancer cell sensitivity and apoptosis.
