Human CRC cells (Caco2 and SW480): AURKA siRNA vs non targeted siRNA

ABSTRACT: Transcriptional profiling of Caco2 and SW480 CRC cells transfected with AURKA siRNA or a control (non-targeted) siRNA. Caco2 and SW480 both have a gain of chromosomal arm 20q, where the AURKA gene is located. The goal was to determine the effects of AURKA knock-down on global gene expression.

Gene expression analysis of SW480 colorectal cancer cells with siRNA mediated ZEB1 or control (GFP) knockdown

Project description:Epithelial-to-mesenchymal transition (EMT) is a fundamental process in development and disease. If aberrantly activated it is a trigger for tumour progression and metastasis (Thiery et al, 2009). It is now known that EMT activation is also associated with the maintenance of stem-cell properties (Mani et al, 2008). Since Zinc-finger enhancer binding transcription factor 1 (ZEB1) is a crucial EMT activator, we analyzed the changes in the gene expression profile that accompany siRNA mediated loss of ZEB1 in SW480 colorectal cancer cells. SW480 is a cell line that exhibits mesenchymal characteristics, but reverts to an epithelial phenotype upon ZEB1 knock down (Spaderna et al. 2006 Gastroenterology). SW480 cells were transfected with control (GFP) or ZEB1 siRNA. 72 hours after transfection, cells were harvested, total RNA was isolated and hybridized.

Identification of genes are regulated by AURKA at transcription level

Project description:Purpose: AURKA plays an important role in breast cancer development. Exploring the gene expression profiles regulated by AURKA will facilitate to understand the mechanism which is responsible for AURKA induced breast cancer development. Results: We found that 350 genes were significantly up-regulated during AURKA overexpression in MCF-10A cells, 346 genes were significantly down-regulated during AURKA overexpression in MCF-10A cells. Conclusions: Our study indicated that 696 differentially expressed genes might contribute to AURKA induced breast cancer development. MCF-10A cells overexpressed AURKA or the empty vector were subjected to RNA extraction. The resulted RNA samples were performed RNA-sequencing analyses of gene expression profiles.

Chromosome 20q amplification regulates in vitro response to Kinesin-5 inhibitor

Project description:We identified gene expression signatures predicting responsiveness to a Kinesin-5 (kinesin spindle protein, KIF11) inhibitor (Kinesin-5i) in cultured colon tumor cell lines. Genes predicting resistance to Kinesin-5i were enriched for those from chromosome 20q, a region of frequent amplification in a number of tumor types. siRNAs targeting genes in this chromosomal region identified AURKA, TPX2 and MYBL2 as genes whose disruption enhances response to Kinesin-5i. Taken together, our results show functional interaction between these genes, and suggest that overexpression of AURKA, TPX2 and MYBL2 is functionally involved in resistance to Kinesin-5i. Furthermore, our results suggest that patients whose tumors overexpress AURKA due to amplifications of 20q will be more likely to resist treatment with Kinesin-5 inhibitor, and that inactivation of AURKA may sensitize these patients to treatment. Project description:The paper describes new experiments about thymosin beta 4 nuclear translocation using different in vitro cellular stress in CaCo2 cell line adding new details regarding the role of this peptide during adaptive and defensive response. To this purpose, CaCo2 cells were submitted to different stress treatments, including serum starvation, DMSO and Butyrate treatment. The expression of Tβ4 was studied in culture cells before and after stress treatment in the cytosolic and nuclear fractions, with the use of different apporaches: immunohistochemistry with anti-Tβ4 antibody; mass-spectrometry characterization and quantification, and mRNA analysis.
