Project description:The endometrium provides optimal conditions for the transport of sperm to the oviduct, to the site of fertilization, and later on for the reception of the embryo. To study these changes on the level of gene expression, a messenger RNA expression profiling of endometrium tissue samples collected from 19 cyclic heifers at five stages of the estrous cycle (days 0, 3.5, 12, 18, 20) was performed. RNA was extracted from these tissue samples and analyzed with a custom-made bovine oviduct and endometrium (BOE) cDNA array. The cDNAs present on the array were derived from several previously conducted differential gene expression studies of bovine endometrium between different stages of the estrous cycle, during early pregnancy, and from studies of bovine oviduct epithelial cells. In all of these studies cDNAs of differentially expressed genes were identified using a combination of subtracted cDNA libraries and cDNA array hybridization. 1,440 cDNA fragments are located on the array. Twenty radioactively labeled cDNA samples (n=4 for each cycle stage) were hybridized with BOE arrays. Raw data were normalized using the BioConductor package vsn. Project description:Here we tested the hypothesis that intrauterine infusion of pathogenic bacteria leads to changes in the transcriptome of the reproductive tract in dairy cattle three months later. We used virgin Holstein heifers to avoid the confounding effects of periparturient problems, metabolic stress and lactation. Animals were infused intrauterine with endometrial pathogenic bacteria Escherichia coli and Trueperella pyogenes to induce clinical endometritis (n = 4) and compared with control animals (n = 6). Three months after infusion, the caruncular and intercaruncular endometrium, isthmus and ampulla of the oviduct, and granulosa cells from dominant ovarian follicles were profiled by RNA-sequencing. Compared with control, bacterial infusion altered the transcriptome of all the tissues. Most differentially expressed genes were tissue-specific, with 109 differentially expressed genes unique to the caruncular endometrium, 57 in the intercaruncular endometrium, 65 in the isthmus, 298 in the ampulla, and 83 in granulosa cells. Surprisingly, despite infusing the bacteria into the uterus, the granulosa cells had more predicted upstream regulators of differentially expressed genes than all the other tissues combined. In conclusion, there was evidence of long-term changes to the transcriptome of the endometrium, oviduct and even the granulosa cells after intrauterine infusion of pathogenic bacteria, which implies that all these tissues contribute to the infertility that persists after endometritis.

Project description:As current bovine pregnancy detection methods are not reliable until at least day 28 post artificial insemination (AI), commercial interest exists for the discovery of novel biomarkers of pregnancy which could reliably detect pregnancy status at or before day 21 of pregnancy. This would allow producers the opportunity to rebreed at the next oestrus event. Therefore, the objective of the present study was to use liquid chromatography tandem mass spectrometry (LC-MS/MS) to perform a global, label-free, proteomics study on (i) whole milk whey and (ii) extracellular vesicle (EV) enriched milk whey samples, from day 21 of pregnancy and day 21 of the oestrous cycle, in order to identify potential protein biomarkers of early pregnancy. The oestrous cycles of 10 dairy cows were synchronised, they went through one (control) oestrous cycle and these cows were artificially inseminated during the following oestrous. These cows were confirmed pregnant by ultrasound scanning. Milk whey samples were collected on day 21 of the oestrous cycle and on day 21 post AI. A Q-Exactive was used to analyse whole milk whey samples and EV enriched milk whey samples by LC-MS/MS and subsequent analyses of the label-free quantitative data was performed in MaxQuant and Perseus. Four proteins (APOB, SPADH1, PLIN2 and LPO) were differentially expressed between the proteomes of whole milk whey from day 21 of pregnancy and day 21 of the oestrous cycle (P<0.05). Ten proteins (PIGR, PGD, QSOX1, MUC1, SRPRA, MD2, GAPDH, FOLR1, GPRC5B and HHIPL2) were differentially expressed between the proteomes of milk whey from day 21 of pregnancy and day 21 of the oestrous cycle (P<0.05). These proteins are potential milk whey biomarkers of early pregnancy; however, the small fold change differences in their abundance between samples obtained during early pregnancy and during oestrous will restrict their use in a pregnancy diagnosis test.

Project description:We compared gene expression in oviduct tissues between unmated (control) and mated hen. As spermatozoa are foreign to the female reproductive tract therefore we were interested to look at how spermatozoa survive in the female reproductive tract and keep their fertilization potentiality. To check that we collected tissues from oviduct of both control and mated female chicken and compared if sperm deposition to the oviduct made any gene expression shift related to sperm survival.

Project description:While the equine oviduct clearly affects early embryo development and while the selective transport of equine embryos through the oviduct indicates a reciprocal interaction, the influence of the embryo on gene expression in the oviduct remains to be determined in the horse. The aim of this study was to examine this by means of RNA sequencing. Four days after ovulation, the oviduct epithelial cells ipsilateral and contralateral to the ovulation side from five cyclic and five pregnant mares were collected. mRNA was extracted and samples were sequenced using the Illumina Hiseq-2000 sequencer. Data analysis was performed with the CLC Genomics software and differentially expressed genes (DEGs) were determined (p-value ≤ 0.05 and absolute fold change ≥ 2). ClueGO was used for functional interpretation. A total of 26,991 genes was identified and 253 genes were found to be upregulated and 108 to be downregulated in the pregnant ipsilateral oviduct, when compared to the cyclic ipsilateral oviduct. Comparison of the ipsilateral and the contralateral oviduct indicated 164 DEGs in pregnant mares and 77 DEGs in cyclic mares. Functional enrichment analysis only detected differences in the comparison of pregnant and cyclic ipsilateral oviducts and showed that the equine embryo affects the expression of immune response related genes in the oviduct, with marked upregulation of interferon associated genes. This research represents the foundation for further assessment of the role of specific genes in the early embryo-maternal dialogue of the horse.
