Gene expression of Myeloid cells stimulated with cytokines.

ABSTRACT: THP-1 is a human monocytic cell line derived from an acute monocytic leukemia patient. It is efficiently converted into macrophages using cytokine combinations and or by using chemical compound Phorbol 12-myristate 13-acetate (PMA). Together with performing gene expression analysis on HSC’s we also performed gene expression analysis on THP-1 cells in order to check which genes have been expressed/overexpressed during differentiation into macrophages. Cells were given treatment with cytokines combination (GMCSF, MCSF). After 24 hours of incubation, cells were washed and pellet was preserved in RNAlater (Ambion) to be shipped to Genotypic Technologies Pvt. Ltd. (Bangalore) for microarray analysis. RNA was isolated by Qiagen’s RNeasy mini kit and was reverse transcribed into cDNA. Cy3 labeled cRNA was formed by cDNA by T7 promoter based linear amplification using Agilent’s Quick amp labeling kit and quantified in Nanodrop Spectrophotometer and their integrity was checked by Bioanalyser. .

Project description:Human CD34+ hematopoietic stem cells gives rise to complete blood lineages through the process of hematopoiesis. Here we tried to differentiate a particular lineage i.e myeloid from CD34+ hematopoietic stem cells using a transcription factor PU.1 and its mutated form. Gene expression analysis was performed after treating cells with protein factors and different cytokine combinations. Cells were given six different treatments including control-PU.1 only and PU.1 with cytokines combination; Y244D only and Y244D with cytokines combination and only cytokine combination (IL3, GMCSF, Flt2, MCSF, and SCF). After 5 days of incubation, cells were washed and pellet was preserved in RNAlater (Ambion) to be shipped to Genotypic Technologies Pvt. Ltd. (Bangalore) for microarray analysis.

Project description:Differentially regulated miRNA candidates in H37Rv infected THP-1 cells were analysed with respect to uninfected THP-1 reference samples. THP-1 cells are monocytes differentiated to macrophages after treatment with PMA for 48 hrs. Total RNA was isolated from infected THP-1 cells after 24 hrs of infection, cDNA was synthesized for TLDA real time PCR reaction using TaqMan MicroRNA Reverse Transcription kit and Megaplex Human Pool A and Pool B stem loop RT primers (version 3.0) as per manufacturer’s protocol. Further real time reaction was performed on QuantStudio 12K Flex Real-Time PCR System (Applied Biosystems) by using cDNA (without pre-amplification) on TLDA card A and card B (version 3.0).

Project description:Resting B cells were isolated from mice splenocytes. Briefly, single cell suspension was prepared and treated with B cell enrichment cocktail supplemented with biotinylated anti-CD43 Abs. The cells were then treated with same volume of streptavidin-magnetic beads and negatively selected on BD IMagnet. The purity of RB cells obtained was >98%, as established by flow cytometry (CD45R^+ CD19^+ CD43^- CD4^- CD8^- ). Purified cells were cross linked with anti-CD86 Abs or anti-CD40 Abs, washed and cultured for 4 hours in the presence or absence of Pam2CSK4. Cells were harvested and washed and treated with RNAlater and subjected to microarray analysis. Microarray was performed by Genotypic Technology Pvt. Ltd. (Bangalore, India; www.genotypic.co.in <http://www.genotypic.co.in/>). The samples for gene expression were labeled using Agilent Quick-Amp labelling kit (p/n5190-0442). The labeled cRNA samples were hybridized on to a Genotypic designed Custom Whole Genome Mouse 8x60k (AMADID No: 26986). Cy3-labeled samples (800 ng) were fragmented and hybridized. Fragmentation of labeled cRNA and hybridization were done using the gene expression hybridization kit of Agilent. Hybridization was carried out in Agilent's Surehyb Chambers at 65 C for 16h. The hybridized slides were washed using Agilent gene expression wash buffers and scanned using the Agilent microarray scanner, G2505C at 3 resolution. Data extraction from images was done using Feature extraction software v 10.5.1.1 of Agilent. Feature extracted data were analyzed using GeneSpring GX v 11 software from Agilent. Genes were classified based on the functional category and pathways using GeneSpringGX Software and Genotypic Biointerpreter-Biological Analysis Software. Bortezomib resistance in THP-1 cell line

Project description:Bortezomib is a proteasome inhibitor used in severel different hematological malignancies. Resistance to this drug is still poorly understood. In order get more insight in the resistance mechanism, we developed several bortezomib resistant subclones of the THP-1 monocytic/macrophage cell line. On these subclones expression arrays were performed. We performed expression array three different bortezomib resistant subclones of the THP-1 cell line. The resistant subclones were spotted against the parental THP-1 wildtype cell line.
