Next generation sequencing based transcriptome comparison between wild-type Drosophila and an Alzheimer's disease model

ABSTRACT: Gene expression profiling of wild-type (WT) fruit flies and transgenic flies that express human amyloid precursor protein was performed to investigate how Aβ42 affects the transcriptome of Drosophila. Overall design: Head tissues from 3-, 10- and 20-day-old male WT and Alzheimer’s disease (AD) Drosophila model were collected for RNA isolation and high-throughput sequencing.

INSTRUMENT(S): HiSeq X Ten (Drosophila melanogaster)

ORGANISM(S): Drosophila Melanogaster

Alzheimer's disease (AD) is an age-related neurodegenerative disease with unknown mechanism that is characterized by the aggregation of abnormal proteins and dysfunction of immune responses. In this study, an integrative approach employing <i>in silico</i> analysis and wet-lab experiment was conducted to estimate the degrees of innate immune system relevant gene expression, neurotoxic Aβ42 generation and neuronal apoptosis in normal <i>Drosophila melanogaster</i> and a transgenic mode  ...[more]
