Transcription profiling of Medicago truncatula time series of flower and pod development

Project description:To study the development of flowers and pods in Medicago truncatula, eight stages of flower and pod development (designated F1-F4 and P1-P4, respectively) were chosen for tissue harvesting. As a common reference, young but already fully developed leaves (corresponding to the third-distant node from the apices) from plants grown in the field were selected. For each stage of flower and pod development, two biological replicates were harvested, one from plants grown in the greenhouse and one from plants grown in the field.

Fundamental molecular bases for differential transcriptional programs of top leaves during grain-filling in rice

Project description:Flag (FL) and second leaves (SL) in rice show differential aging patterns during monocarpic senescence. Coordination of aging programs in the top leaves is important for effective grain-filling. However, molecular bases for differential aging programs in the top leaves have not been systematically explored in rice. Here, we performed mRNA-sequencing of FL and SL at six time points during the grain-filling period. mRNA expression data revealed 6,365 genes showing aging-dependent expression changes in FL and/or SL. Of them, while 3047 genes showed shared aging-dependent expression patterns between FL and SL, 3058 genes showed differential expression patterns, which were classified into 5 major groups (G1-5) based on their differential expression patterns. Of the groups, G3 representing amino acid (AA) transport showed consistent differential age-dependent expression patterns in independent samples, whereas the other groups showed inconsistent differential expression patterns. Moreover, of AA transporters (AATs) in G3, long-distance AATs showed invariant differential age-dependent expression patterns after panicle removal, consistent to panicle removal-invariant differential nitrogen contents between FL and SL, known to be associated with protein concentration in grains. Our results suggest that long-distance AA transport is an invariant core transcriptional program of differential aging in rice top leaves for nitrogen remobilization during grain-filling.

Samples were collected from Well Watered (WW) plants at the beginning of the stress imposition (point A, T=0), and from Water-Stressed (WS) and WW control plants at the end of the drought period (point B, T=+8). Samples named SEED consisted of seeds from the pod of the second flowering node (seed-WW-A, seed-WW-B and Seed-WS-B). Samples named LEAF consisted of the leaves and stem sections from the two vegetative nodes below the first flowering node (leaf-WW-A, Leaf-WW-B and Leaf-WS-B). Each sample consited of a pool of 3 individual plants and 4 repetitions per condition were carried out.

A transcript profile of Medicago truncatula 2HA tissue culture with auxin treatment

Project description:Leaf explants of the superembryogenic Medicago truncatula line 2HA were treated with auxin (1-naphthaleneacetic acid) for one week to induce the formation of roots (Imin et al J Exp Bot 58:439-451). Gene expression in the leaves and the NAA treated tissue cultures was compared to identify transcripts expressed during the commitment to root formation in tissue culture. We have used the Affymetrix Medicago Genome Array GeneChip to compared gene expression in Medicago truncatula leaves and leaf explants that have been cultured for one week on NAA, to identify genes expressed during the commitment to root formation in tissue culture. MtIFS1-expressing alfalfa

Project description:We used an M. truncatula 16K oligonucleotide array to profile gene expression in the leaves of transgenic alfalfa plants expressing Medicago truncatula isoflavone synthase 1 (MtIFS1). RNA purified from leaves of MtIFS1-expressing lines C22 and B20 and vector control line VC11 was used to generate Cy5-labeled cDNA. RNA purified from a second vector control line, VB2, was used to generate Cy3-labeled reference cDNA. Three independently propagated cuttings of each line were used and a total of nine hybridizations were performed.
