Gene expression profiling of IRBIT mutant flies

ABSTRACT: In order to understand the biological role of Drosophila IRBIT, we generated null mutant fruitflies. We performed Illumina platform-based RNA-Seq analysis to investigate transcriptional consequences of the mutation using fly midguts as well as whole animals.

Expression profile of third instar larvae tissues from wild type and bsAS mutant flies

Project description:Our data on third instar larvae tissues of Drosophila melanogaster has allowed us to identify bsAS, a lncRNA antisense to the blistered/DSRF gene, orthologous to the SRF transcription factor in humans, mainly expressed in fly wings. bsAS controls the expression of different isoforms of the blistered gene in Drosophila tissues. CRISPR-CAS deletion of bsAS Transcription Start Site thoroughly abolishes bsAS transcription, inducing a change in the bs isoform usage and the impairment of wing development. Our data consists on stranded RNA-Seq from eye-antenna, leg and wing imaginal discs from third instar larvae. To analyse bsAS mutation, we also performed stranded RNA-Seq of wild type and bsAS-/- eyes and wings from third instar larvae and late pupae.

Characterization of the effect of hepatocyte nuclear factor 4 (Hnf4) silencing in enterocytes on gene expression in the developing Drosophila larval midgut

Project description:The main objective of this study was to characterize the effects of Hnf4 silencing in enterocytes (ECs) on the Drosophila midgut during larval development. Larval midguts from control animals and animals expressing Hnf4 RNAi in ECs were collected on day three and four after egg laying, corresponding to the late L2 and early L3 stages, respectively. Midguts were then processed for RNA extraction and mRNA sequencing. Our mRNA-seq study reveals significant changes in the transcriptome of control midguts between days three and four of larval development, with increased expression of numerous genes involved in lipid metabolism. Silencing Hnf4 largely suppresses this transcriptional switch, highlighting the role of this nuclear receptor in inducing intestinal lipid metabolism during larval development.

Project description:Differentiated cells exhibit cellular plasticity,where they dedifferentiate into stem cells. We studied cellular plasticity of secretory enternendocrine cells (EEs) in the fruit fly Drosophila melanogaster. We used flies that had EEs lineage-traced and resolved single-cell transcriptomics of all cell types in their midguts. Our scRNA-seq data have characterized cell-type-gene expression profiles, with a specific focus on EE lineages.

Comparative microarray analysis of psh and Dif mutant flies treated with Metarhizium anisopliae ARSEF 549

Project description:The Toll pathway is the chief antifungal pathway in Drosophila. Two components of this pathway have disparate effects on Drosophila infected with Metarhizium ansiopliae ARSEF 549 (Ma549). Infected Drosophila Dif mutants show no change in susceptibility while infected psh mutants rapidly succumb to infection. We evaluated the impact of Ma549 on gene regulation in flies deficient in these Toll pathway components to identify the cause for these susceptibility differences. Four fly lines were used: psh, BDSC6326 (psh control), Dif, and cn bw (Dif control). RNA was collected 46 hrs post infection for 10 infected or uninfected male flies per replicate.
