Gene expression in chicken embryo liver

ABSTRACT: We have profiled the gene expression in eight chicken embryo livers in the 11th day of incubation. Four eggs (labelled as WAT1-4) were injected with 0.15 ml/egg deionized water in days 9, 10 an 11 post incubation while four eggs (labeled CTR1-4) were kept as environmental controls. We found that water injections had a very small effect, regulating less than 1% of the genes. Both WAT and CTR data sets were used as references in further experiments for eggs expossed to various water-soluble carcinogenic compounds.

The eggs were injected on postincubation days 9, 10 and 11 with 4mg activation-dependent DNA reactive diethylnitrosamine (DEN) or its structural non-carcinogenic comparator, N-nitrosodiethanolamine (NDELA)DEN in 0.15 ml/egg deionized water. We found that both compounds induced simmilar massive transcriptomic alterations in the xenobioatic metabolism pathways when comparing with reference data in eggs injected with the vehicle only (deionized water).

Phenological transcriptomic profile of double-crested cormorants during embryonic development

Project description:In this study, we tracked the phenology of the transcriptome during double-crested cormorant (Nannopterum auritum) embryogenesis. Fresh eggs were collected from a reference site and artificially incubated from collection until four days prior to hatching. Embryos were periodically sampled throughout incubation for a total of seven time points. A custom microarray was designed for cormorants (over 14,000 probes) and used for transcriptome analysis in whole body (days 5, 8) and liver tissue (days 12, 14, 16, 20, 24). Three main development periods (early, mid, and late incubation) were identified with differentially expressed genes, gene sets, and pathways within and between each developmental transition.

Comparative proteomics analysis of soybean sprouts that treated with deionized water or with 6 mM Ca2+

Project description:Soybean sprout, a kind of year-round vegetable in Asia, is perceived as a part of a healthy diet. We found that supplemental Ca2+ could increase soybean sprout yield and improve its nutrition qualities. Ca2+-treated sprouts had higher yield than water-treated ones. Metabolism of selected anti-nutritional factors and bioactive substances in soybean sprouts was strengthened by Ca2+. To investigate the role of Ca2+ in soybean during germination, proteomic changes were analyzed. Total protein from soybean sprouts that treated with deionized water or with 6 mM Ca2+ were analyzed.

The effects of phthalates on S. tropicalis development

Project description:Phthalates are added to polymers in order to increase their flexibility, but leach out of the matrix. Given that these plasticizers are mainly found in water bound to particles, aquatic species are exposed to phthalates. Silurana tropicalis eggs were exposed to high concentrations of three pththalates: dicyclohexyl phthalate (DCHP; 19 mg/L), dimethyl phthalate (DMP; 34 mg/L), and monomethyl phthalate (MMP; 1500 mg/L), along to their respective controls: 0.82% DMSO for DCHP, and water controls for DMP and MMP. The exposure lasted 3 days during embryogenesis. Four pooled of 10 larvae were sampled per treatment and analyzed for gene expression changes using a microarray approach.

The majority of Cu is taken up into the cell through the Cu transporter CTR1 and thus, deletion of Ctr1 results in Cu deficiency. During an indirect life cycle, immature eggs are passed out of the host in the stool and embyonate in the external environment until the release of miracidium which invades a freshwater snail - intermediate host. By utilization of RNA-seq technique (Illumina NextSeq 500), differentially expressed genes were analysed in F. hepatica eggs during the maturation process in three time points: 0 days (freshly laid eggs by adult F. hepatica individuals) and after incubation for 5 and 10 days.
