Inhibition of embryonic HSP-90 function causes variation of cold tolerance in zebrafish

ABSTRACT: Accumulating evidence suggests that HSP90 plays roles in modulation of phenotype in vertebrate development, which is closely involved in adaptation and evolution. In this study, we showed that inhibition of embryonic HSP90 function, either by a chemical inhibitor （radicicol） or low conductivity stress, induced variation of cold tolerance in adult zebrafish.

Project description:We wanted to study regulation of gene regulation by sba1 (ortholog of mammalian p23) in yeast. sba1/p23 binds to Hsp90 and acts as a co-chaperone. We have a small molecule, radicicol, an analog of geldanamycin to inhibit the binding of p23 to Hsp90. Consequently, we have analyzed the gene expression profile in 4 conditions, wt, delta-sba1; and when Hsp90 function is impaired, that is, wt and delta-sba1 treated with radicicol.

Project description:Aneuploidy, a state of having uneven numbers of chromosomes, is a form of large-effect mutation able to confer adaptive phenotypes under diverse stress conditions. Here we investigate whether pleiotropic stresses could in turn induce aneuploidy in budding yeast. Cells exposed to radicicol, an Hsp90 inhibitor, exhibit chromosomal instability. To assess the karyotype of cells with an acquired resistance to radicicol, array CGH was used to compare three stably resistant yeast colonies to a WT strain. Single replicates of DNA prepared from radicicol-resistant yeast colonies are compared to a control WT colony.

Project description:In this study we provide evidence that Hsp90 binds chromatin at specific sites close to several TSS in Drosophila S2 cell line. In addition of finding a preference for stalled promoter regions of annotated genes, we uncover many intergenic Hsp90 binding sites coinciding with non-annotated transcription start sites. Interestingly, this set includes promoters for primary transcripts of microRNA genes, thereby expanding the scope of Hsp90 to transcriptional control of many genes. We finally conclude that Hsp90 contacts NelfE and thus regulates pol II pausing.
