Project description:The purpose of this study is to obtain comprehensive gene expression profiles in breast cancer. Mammary gland cells were specifically isolated from 433 clinical tissue samples by laser capture microdissection (LCM). Total RNAs were extracted from LCM captured samples. We investigated gene expression profiles in 417 patients with breast cancer and 16 non-tumor tissues as a normal control using an Affymetrix GeneChip. Mammary gland cells were captured from clinical tissues of breast cancer patient by LCM. Gene expression profilings for 417 tumor and 16 non-tumor samples were acquired using GeneChip Human Genome U133 Plus 2.0 (Affymetrix, Santa Clara, CA). Microarray datasets were normalized and transformed to log2 values using robust multi-array average (RMA) method with R statistical software and BioConductor package.

Project description:EPR is a long non-coding RNA (lncRNA) that controls cell proliferation in mammary gland cells by regulating gene transcription. Here, we report on Mettl7a1 as a direct target of EPR. We show that EPR induces Mettl7a1 transcription by rewiring three-dimensional chromatin interactions at the Mettl7a1 locus. Our data indicate that METTL7A1 contributes to EPR-dependent inhibition of TGF-β signaling. METTL7A1 is absent in tumorigenic murine mammary gland cells and its human ortholog (METTL7A) is downregulated in breast cancers. Importantly, re-expression of METTL7A1 in 4T1 tumorigenic cells attenuates their transformation potential, with the putative methyltransferase activity of METTL7A1 being dispensable for its biological functions. We found that METTL7A1 localizes in the cytoplasm whereby it interacts with factors implicated in the early steps of mRNA translation, associates with ribosomes, and affects the levels of target proteins without altering mRNA abundance. Overall, our data indicates that METTL7A1 —a transcriptional target of EPR— modulates translation of select transcripts.

Project description:In this study, two small RNA libraries were constructed using dry period and peak lactation dairy goat mammary gland tissues and sequenced by the Illumina Solexa high-throughput sequencing system. A total of 346 conserved and 95 novel miRNAs were identified in the dairy goat. The expression of miRNAs was confirmed by qRT-PCR in nine tissues and the mammary gland during development cycles. In addition, several candidate miRNAs that may be involved in mammary gland development and lactation were found by the comparison of miRNA expression profiles among different tissue and developmental stages of the mammary gland. This study provides the identification and profile of miRNAs related to the biology of the mammary gland in the dairy goat. The identification of these miRNAs could contribute to understanding the molecular mechanisms of lactation physiology and the development of the mammary gland in the dairy goat.

Project description:Mycoplasma bovis (M. bovis) causes to the diseases such as arthritis, pneumonia, abortion and mastitis causing to great losses in dairy bovine industries. It undertakes significant functions in the regulation of the immune response formed by many RNA type bacteria such as Messenger RNA (mRNAs) and Long noncoding RNA (lncRNAs). mRNA and lncRNA expression profiles still cannot be majorly known in the bovine mammary gland tissues infected with M. bovis. To illuminate this issue, transcription analysis has been conducted regarding mRNA and IncRNAs in the breast tissues belonging to Holstein cattle infected and not infected with M. Bovis. Differently expressed 1310 mRNAs and 57 IncRNAs have been determined in the bovine mammary gland tissues infected and not infected with M. Bovis. In addition; 392 novel lncRNAs have been detected. Also; it has been found that 19 novel lncRNAs are expressed in a different way. It has been revealed in gene ontology analysis that the differently expressed mRNAs and IncRNAs play a significant role in many vital biological pathways such as Metabolic pathways, T cell receptor signaling pathway, TGF-beta signaling pathway, Pathways in cancer, PI3K-Akt signaling pathway, NF-kappa B signaling pathway, mTOR signaling pathway and Apoptosis including the immune response and cancer. When literature review is conducted, it is seen that this study is the first genome wide IncRNA research conducted on the infected bovine mammary gland tissues with M. bovis agent. Our results have provided bovine mammary gland IncRNA and mRNA resource to understand their roles in the regulation of the immune response against the agent of M. Bovis in the bovine mammary gland tissues.

Project description:Genome-wide analyses have identified thousands of long non-coding RNAs (lncRNAs). Malat1 (Metastasis Associated Lung Adenocarcinoma Transcript 1) is among the most abundant lncRNAs whose expression is altered in numerous cancers. Here we report that genomic loss, as well as systemic knockdown of Malat1 using antisense oligonucleotides, in the MMTV-PyMT mouse mammary carcinoma model results in slower tumor growth accompanied by differentiation into highly cystic tumors and a significant reduction in lung metastasis. Further, Malat1 loss results in a reduction of branching morphogenesis in MMTV-PyMT and Her2/neu amplified tumor organoids consistent with the in vivo reduction in lung metastasis. At the molecular level, Malat1 knockdown results in alterations in gene expression and changes in splicing patterns of genes involved in differentiation and pro-tumorigenic signaling pathways. Together, these data indicate that the lncRNA Malat1 regulates critical processes in mammary cancer pathogenesis and represents a promising therapeutic target for inhibiting breast cancer metastasis.
