Project description:Aim of this experiment was to characterize whether SPP1 released from the hippocampal perivascular space could imprint microglia transcriptional states. We performed scRNA-seq analysis on sorted CD140a+ perivascular fibroblast (PVF), CD45highCD11intCD206+ perivascular macrophage (PVM) and CD45intCD11intCX3CR1high microglial cell from dissected hippocampi of 6-month old wild type vs SPP1KO/KO vs AppNL-F mice vs AppNL-FxSpp1KO/KO mice. Cells were isolated from dissected hippocampi as previously described (Sala Frigerio et al.,2019).

Project description:Fear conditioning induces immediate changes in the mouse hippocampal transcriptome profile. Here, we performed RNA sequqencing in mice hippocampi at 15 minutes, 1 hour and 3 hours post-fear conditioning in addition to non-conditioned mice. Tob gene has been shown to affect cellular stress and behavior. In order to understand the role of Tob gene in the hippocampal stress and fear machinery, we compared hippocampi of Tob WT and KO mice. This dataset shows the temporal transcriptomic changes in mouse hippocampus induced by fear conditioning. Also, it shows the changes occurred after Tob deletion.

Project description:MicroRNAs (miRNAs) are small endogenous regulatory RNAs involved in hippocampal functions and lesions. Mouse has been regarded as an effective tool to explore the roles of miRNAs in hippocampus. Although the anatomical structure and physiological function across human and mouse hippocampi are highly conserved, the similarity and difference of hippocampal miRNAomes between the two species have not been well characterized. Our present study represents the first attempt to perform a systematic comparison of the miRNAomes between healthy human and mouse hippocampi using high-throughput sequencing followed by bioinformatic analyses. Contrasting between human and mouse hippocampal miRNAs shows a global similarity of expression pattern; however, the interspecific expression conservation of hippocampal miRNAs is positively associated with the interspecific sequence conservation. Although the type of the abundantly expressed hippocampal miRNAs is almost the same across the two species, only the enriched miRNAs in human hippocampus exhibit potential neuro-related activities. In addition, we also identified a novel miRNA and a species-specific miRNA in human hippocampus whose putative targets are associated with neuro-related activities. Collectively, our results can provide a framework to explore the ability of mice to model the physiological and pathological processes of human hippocampus and also aid in understanding the basis for higher order abilities of learning and memory at the miRNA level.

Project description:Gene expression profiles of Ngn3-overexpressing cultured hippocampal neurons was compared to the profile of the corresponding control populations. (neurons expressing GFP). Neurogenin3, a proneural transcription factor controlled by Notch receptor, is involved in hippocampal neuron differentiation and synapses, but little is known about the molecular bases of Ngn3 activity in neurons. Microarray analysis indicated that overexpression of Ngn3 upregulated a number of genes related with cytoskeleton dynamics. One of then was Fmn1 whose protein is associated with actin and microtubule cytoskeleton. Overexpression of the isoform Fmn1-Ib in cultured hippocampal neurons induced an increase in the number of primary dendrites and in the number of glutamatergic synaptic inputs without affecting GABAergic synapses resulting in a modification in the balance between excitation and inhibition. The same changes were provoked by overexpression of Ngn3. In addition downregulation of Fmn1 by the use of Fmn1-siRNAs impaired such morphological and synaptic changes induced by Ngn3 overexpression in neurons. These results reveal a previously unknown involvement of Formin1 in dendritic and synaptic plasticity as a key protein in the Nng3 signaling pathway that contributes to understanding of molecular mechanisms of the neuronal differentiation. Project description:The hippocampus is important for memory formation and is severely affected in the brain with Alzheimer's disease (AD). Since AD brain tissue is available postmortem, our understanding of early pathogenic processes occurring in hippocampi remains speculative. Here, an MS-based proteomic approach was used to analyze free-floating hippocampal spheroids (HSs) enriched in PROX1-positive granule neurons, from induced pluripotent stem cells (iPSCs) of healthy individuals and AD patients. Project description:Calorie restriction (CR) enhances longevity and mitigates aging phenotypes in numerous species. Physiological responses to CR are cell-type specific and variable throughout the lifespan; however, the mosaic of molecular changes responsible CR benefits remain unclear, particularly in brain regions susceptible to deterioration throughout aging. Thus, we examined the influence of long-term CR on the CA1 hippocampal region, a key learning and memory brain area that is vulnerable to age-related pathologies, such as Alzheimerâs disease (AD). Through mRNA sequencing and NanoString nCounter analysis, we demonstrate that one year of CR feeding suppresses an age-dependent signature of 882 genes functionally associated with synaptic transmission-related pathways, including calcium signaling, long-term potentiation (LTP), and Creb signaling in wild-type mice. By comparing the influence of CR on hippocampal CA1 region transcriptional profiles at younger- (5 months) and older-adult (15 months) timepoints, we identify conserved upregulation of proteome quality control and calcium buffering genes, including heat shock 70 kDa proteins 1b and 5 (Hspa1b and Hspa5), protein disulfide isomerase family A members 4 and 6 (Pdia4 and Pdia6), and calreticulin (Calr). Expression levels of putative neuroprotective factors, klotho (Kl) and transthyretin (Ttr), are also elevated by CR throughout adulthood, although the global CR-specific expression profiles at young and older timepoints are highly divergent. At a previously unachieved resolution, our results demonstrate conserved activation of neuroprotective gene signatures and broad CR-suppression of age-dependent hippocampal CA1 region expression changes, indicating that CR functionally maintains a more youthful transcriptional state within hippocampal CA1 throughout aging. Hippocampal CA1 region mRNA profiles of younger- (5 months) and older-adult (15 months) mice on calorie-restricted (CR) and normal (AD) diets were generated by deep sequencing using Illumina HiSeq 2500.

Project description:Neural stem cells (NSCs) generate new neurons throughout life in two distinct areas of the mammalian brain: the subventricular zone (SVZ) lining the lateral ventricles and the hippocampal dentate gyrus (DG). How gene expression signatures differ among NSCs and immature neurons within and between these adult neurogenic regions is unknown. We isolated NSCs and their progeny using transgenic mice expressing GFP under the control of the Sox2 promoter (labeling NSCs) and transgenic mice expressing DsRed under the control of the doublecortin (Dcx) promoter (labeling immature neurons). Comparison of the transcriptomes of SOX2+ cells derived from both neurogenic areas revealed that NSCs are highly similar but that functionally significant differences in gene expression exist: IGF2, which is expressed only in SOX2+ cells in the DG but not in the SVZ, is required for proliferation of DG-derived but not SVZ-derived NSCs. Gene expression profiles strongly diverged in immature neurons, and we provide evidence that ephrinB3, which was up-regulated only in the DG but not in the SVZ during neuronal differentiation, regulates the survival of newborn granule cells. Thus, the data provided here show that stem cell populations in the adult DG and SVZ are similar but have unique properties that manifest themselves later during neural differentiation, resulting in distinct neuronal populations Hippocampi and SVZ from 6 week old DCX-DsRed and Sox2-GFP Reporter mice were dissected and cell sorted using FACS. cDNA were generated and analysed using Agilent Platform.
